Gut microbiota-derived bile acids in intestinal immunity, inflammation, and tumorigenesis

Jie Cai et al.

Summary: Inflammatory bowel disease (IBD) and colorectal cancer (CRC) are diverse intestinal diseases threatening an increasing number of individuals as their lifestyles become westernized. Recent research has found that gut microbiota-derived metabolites play a crucial role in maintaining intestinal homeostasis and modulating the progression of intestinal diseases.

Alcohol triggered bile acid disequilibrium by suppressing BSEP to sustain hepatocellular carcinoma progression

Wenbo Chen et al.

Summary: This study found that alcohol consumption could disturb the homeostasis of bile acids (BAs) and promote hepatocellular carcinoma (HCC) progression by activating oncogenes and suppressing tumor suppressor genes. The study also revealed a decrease in BSEP and an increase in certain histone methyltransferases in alcohol-exposed liver cancer cells, indicating the impact of alcohol on BAs homeostasis.

Comparison of bile acids profiles in the enterohepatic circulation system of mice and rats

Zhicheng Duan et al.

Summary: Bile acids are biomarkers for liver diseases and play a role in liver-gut communication. The homeostasis of bile acids is controlled by multiple factors and shows significant species differences. Our study reveals distinct differences in bile acid profiles between rats and mice, especially in the intestine.

A convenient and efficient 4-(diethylamino)-butylamine-lab ele d polarity-response-homodispersed strategy for absolute quantification of carboxyl submetabolome: Monitoring the whole progressive course of hepatocellular carcinoma

Minghui Tong et al.

Summary: Recent studies have shown that dysregulation of carboxyl submetabolome homeostasis is closely related to the occurrence and development of hepatocellular carcinoma (HCC). This study established a convenient and efficient workflow for absolute quantification of carboxyl submetabolome, which was applied to monitor carboxyl metabolic profile in human plasma of the whole progressive course of HCC. Biomarkers of health-hepatitis-cirrhosis-HCC were screened out and a diagnostic model was established. The relevancy between carboxyl submetabolome disorders and the pathogenesis of HCC was also investigated.

Summary: This study investigated the physiological function of BAAT in bile acid and lipid metabolism using Baat(-/-) mice. The results showed that BAAT deletion affected bile acid synthesis and metabolism, altered energy metabolism and microbiome structure in mice. Furthermore, when fed a westernized diet, Baat(-/-) mice showed compromised weight gain and impaired insulin secretion.

Bile acids as regulatory molecules and potential targets in metabolic diseases

Ai-jin Xie et al.

Summary: Bile acids, derived from cholesterol in the liver, play crucial roles in intestinal lipid absorption and various physiological functions. Some bile acids have therapeutic potential, but accumulation of toxic bile acids can cause cell damage. Changes in bile acid metabolism and interactions with gut microbiota are key factors in the pathogenesis of metabolic diseases.

Summary: This review discusses the risk factors, global trends, and high incidence rates of HCC in Asia and Africa. Interventions to minimize the global burden of HCC include reducing common risk factors, treating underlying causes effectively, and implementing comprehensive screening programs for HCC. Healthcare inequity is identified as a crucial area for improvement in addressing disparities in HCC incidence and mortality among racial and ethnic minorities in multiethnic countries.

DEN-Induced Rat Model Reproduces Key Features of Human Hepatocellular Carcinoma

Keerthi Kurma et al.

Summary: Hepatocellular carcinoma (HCC) is the most common type of liver cancer, often associated with liver fibrosis or cirrhosis from chronic liver injuries. The liver's immune system plays a crucial role in disease progression towards HCC. The DEN-induced rat model of HCC exhibits molecular characteristics similar to human HCC, especially those with high proliferation, making it a valuable tool for preclinical testing.

Summary: Over 90% of hepatocellular carcinoma cases occur in patients with cirrhosis, with hepatitis B virus and hepatitis C virus as leading causes. Immunotherapy has revolutionized HCC treatment by targeting immune cells rather than cancer cells, particularly focusing on regulatory T cells for their role in tumor progression and response to immune checkpoint inhibitors.
