Cell and Tissue Nanomechanics: From Early Development to Carcinogenesis

Summary: The study of cell and tissue nanomechanics is important for understanding the characteristics of normal and diseased tissues, as well as the mechanisms of pathological processes. There are similarities between early development and carcinogenesis. Changes in the mechanical properties of cells and their microenvironment occur during carcinogenesis, with a rearrangement of the tissue skeleton involving the extracellular matrix, cytoskeleton, and nuclear scaffold. These changes are associated with the cancer stem-cell niche and offer potential for new therapeutic strategies.

Single-nucleus transcriptomics of IDH1-and TP53-mutant glioma stem cells displays diversified commitment on invasive cancer progenitors

Summary: This study characterized rare subpopulations of glioma stem cells (GSCs) with IDH1 and TP53 mutations using single-nucleus transcriptomics. It was found that double-mutant GSCs showed a commitment to highly invasive oligodendrocyte- and astroglia-like progenitors. These GSCs also exhibited upregulated markers related to collagen synthesis, altered lipogenesis, and high migration. The findings suggest potential treatment targets for different subtypes of gliomas.

Association of glioma CD44 expression with glial dynamics in the tumour microenvironment and patient prognosis

Summary: This study identified CD44 as an independent prognostic factor for lower-grade gliomas (LGGs) by analyzing the ratio of immune and stromal components in the tumor microenvironment (TME). The research also revealed that astrocytes enhance the viability and migration of glioma cells by upregulating CD44 expression, and CD44 expression is positively correlated with M1 macrophages. These findings highlight the important role of CD44 in the TME and suggest it as a potential therapeutic target and prognostic marker for LGGs.

CD44: A Multifunctional Mediator of Cancer Progression

Summary: CD44, a non-kinase cell surface transmembrane glycoprotein, is widely implicated as a cancer stem cell marker in multiple cancers. Cells overexpressing CD44 possess CSC traits and resistance to therapy. The CD44 gene undergoes alternative splicing, resulting in different isoforms that interact with various ligands and drive cancer-associated signaling pathways. However, the association between CD44 expression levels and clinicopathological features is contradictory, although high CD44 expression significantly contributes to tumorigenic mechanisms.
