LRRK2 and idiopathic Parkinson's disease

Emily M. Rocha et al.

Summary: The etiology of idiopathic Parkinson's disease involves multiple factors, including genetics and environmental exposures. LRRK2 plays an important role in iPD, regardless of mutations, by influencing kinase activation and substrate phosphorylation, which contribute to the pathogenesis.

Preclinical and clinical evaluation of the LRRK2 inhibitor DNL201 for Parkinson's disease

Danna Jennings et al.

Summary: Mutations in LRRK2 are common genetic risk factors for Parkinson's disease. Inhibition of LRRK2 with DNL201 has shown positive effects in preclinical and clinical trials, improving lysosomal function and demonstrating safety and tolerability. Further development of LRRK2 inhibitors as a therapeutic modality for Parkinson's disease is warranted.

LRRK2 protects immune cells against erastin-induced ferroptosis

Asmaa Oun et al.

Summary: This study investigates the impact of LRRK2 on ferroptosis-induced cell death in immune cells. The results suggest a protective role of LRRK2 against erastin-induced ferroptosis through modulation of cellular metabolism and mitochondrial function.

Curcumin Reduced H2O2- and G2385R-LRRK2-Induced Neurodegeneration

Jinru Zhang et al.

Summary: Mutations in the LRRK2 gene are common genetic factors contributing to Parkinson's disease, especially the G2385R-LRRK2 mutation in the Chinese population. This study demonstrates that G2385R-LRRK2 induces neurotoxicity through mitochondrial ROS increase and enzyme activation. Additionally, the antioxidant curcumin shows potential in protecting against G2385R-LRRK2-induced neurodegeneration and environmental stressors can exacerbate the neurotoxic effects of both wild-type and mutant LRRK2.
