Summary: Extensive research has been conducted to understand the molecular and regulatory mechanisms involved in phytochrome-mediated light signaling in plants. Downstream signaling components that interact with phytochromes, including negative transcription factors (PIFs) and an E3 ligase complex (COP1 and SPAs), have been identified. The autophosphorylating protein kinase activity of phytochrome A (phyA) has been shown to be important in regulating plant photoresponses.

Understanding the role of PIFs in crop improvement could lead to the development of climate-tolerant crops and increased crop yield.

Additionally, there may be functional interaction between phyA and the defense-related hormone jasmonic acid.

Novel and multifaceted regulations of photoperiodic flowering by phytochrome A in soybean

Summary: Photoperiod is an important environmental cue that allows plants to control the timing of flowering. Soybean, a short-day crop, exhibits variations in flowering time at different latitudes, affecting yields. This study reveals the regulatory mechanism of phyA2 and phyA3 in soybean, showing that they directly bind to LUX proteins to regulate E1 stability, thereby controlling the core flowering suppressor E1 at both the transcriptional and posttranscriptional levels.

Suppression of Phytochrome-Interacting Factors Enhances Photoresponses of Seedlings and Delays Flowering With Increased Plant Height in Brachypodium distachyon

Summary: This study investigated the functions of BdPIL1 and BdPIL3, two PIF-like genes in Brachypodium distachyon, and found that they interact with phytochromes in an active form-specific manner, affecting plant growth and development. Transgenic Brachypodium seedlings with RNAi constructs of BdPIL1 and BdPIL3 showed decreased coleoptile lengths, increased leaf growth, taller stature, and delayed flowering compared to wild-type plants, with downregulation of genes related to cell number regulation, floral induction, and chlorophyll biosynthesis. Additionally, the study revealed the DNA-binding ability of BdPIL1 and BdPIL3 to target promoters was inhibited in the presence of phytochromes, providing insights into the molecular mechanism underlying phytochrome-mediated PIF regulation in Brachypodium.

Out of the Dark and Into the Light: A New View of Phytochrome Photobodies

Photobodies, which are subnuclear compartments formed by phytochromes in response to red light, regulate phytochrome-mediated signaling and physiological outputs. The exact function, composition, and biogenesis of photobodies remain unclear, but they have been suggested to undergo liquid-liquid phase separation based on cellular dynamics and internal components. Understanding these processes can potentially help improve sunlight capture and the fitness of important crops.
