Implicit-Solvent Coarse-Grained Simulations of Linear-Dendritic Block Copolymer Micelles

Summary: The design of nanoassemblies can be achieved by adjusting the strength of hydrophobic interactions in block copolymers. These supramolecular aggregates have attracted attention in drug delivery and imaging due to their tunable properties and scalable production. In this study, we investigated the micellization process of linear-dendritic block copolymers to understand the effect of branching on micellar properties. We found that excluded volume interactions strongly influence the aggregation and charge of micelles at higher degrees of branching and for shorter polymer chains.

Hybrid Molecules Consisting of Lysine Dendrons with Several Hydrophobic Tails: A SCF Study of Self-Assembling

Summary: In this article, the micellization of hybrid molecules consisting of one polylysine dendron with charged end groups and several linear hydrophobic tails attached to its root was studied using the numerical self-consistent field method of Scheutjens-Fleer. The focus was on the formation of spherical micelles and the determination of the parameters for their appearance. The relationship between the size and structure of spherical micelles and the length and number of hydrophobic tails, as well as the number of dendron generations, was established. The relationship between the structure of hybrid molecules and the electrostatic properties of the resulting micelles was also investigated.

NMR Studies of Two Lysine Based Dendrimers with Insertion of Similar Histidine-Arginine and Arginine-Histidine Spacers Having Different Properties for Application in Drug Delivery

Summary: This paper studies two lysine-based peptide dendrimers with Lys-His-Arg and Lys-Arg-His repeating units and terminal lysine groups. The combination of histidine and arginine in these dendrimers could be important for biomedical applications, such as preventing dendrimer aggregation and facilitating dendrimer penetration through cell membranes. The synthesis of these dendrimers is described, and their structure is confirmed using 1D and 2D Nuclear Magnetic Resonance (NMR) spectroscopy. NMR spectroscopy and relaxation are used to investigate the structural and dynamic properties of these dendrimers and compare them with previously studied dendrimers with Lys-2Arg and Lys-2His repeating units. The results show that both Lys-His-Arg and Lys-Arg-His dendrimers exhibit pH-sensitive conformation and dynamics. The Lys-His-Arg dendrimer, which has a similar conformation to the more hydrophobic Lys-2His dendrimer, could be a more suitable candidate as a pH-sensitive nanocontainer for drug delivery compared to Lys-2His and Lys-Arg-His dendrimers.

Enhanced endosomal escape of dendrigraft poly-L-lysine polymers for the efficient gene therapy of breast cancer

Summary: A surface modification strategy was used to enhance the endosomal escape ability of dendrimer DGL polymers, improving gene transfection efficiency. This strategy involved designing a library of PBA-modified DGL polymers to screen for efficient siRNA vectors, with one candidate showing nearly 90% gene silencing in cells. Furthermore, the PBA modification not only improved siRNA cellular uptake but also endowed DGL polymers with the ability to escape endosomes.

Summary: Reticulating PEG with DGL can provide intrinsic cell functionalities to PEG-based hydrogels and tailor the mechanical properties of the hydrogels. Cell adhesion is influenced by the concentration of DGL, cell viability depends on the ratio of DGL and PEG, and cell morphology and proliferation are mainly related to the hydrogel rigidity.

Local Orientational Mobility of Collapsed Dendrimers

Summary: This study investigated the orientational mobility of collapsed and swollen dendrimers using NMR relaxation experiments and atomistic simulations, revealing that volume interactions suppress internal local mobility while reorientation occurs due to the rotation of the dendrimer as a whole. These findings suggest that considering segmental mobility is crucial for interpreting the mobility of CH2 groups observed in NMR experiments.

Size and Structure of Empty and Filled Nanocontainer Based on Peptide Dendrimer with Histidine Spacers at Different pH

Summary: This study compared the properties of dendrimers at two different pH values through molecular dynamics simulation, revealing that dendrimers at low pH have larger size and size fluctuations. The electrostatic properties of dendrimers differ, but are in line with the theoretical soft sphere model and are practically temperature-independent. The effect of inserting histidine residues on dendrimers is stronger in dendrimers with neutral histidines compared to dendrimers with protonated histidines.

Evaluation of transgene expression characteristics and DNA vaccination against melanoma metastasis of an intravenously injected ternary complex with biodegradable dendrigraft poly-L-lysine in mice

Summary: A biocompatible splenic vector for a DNA vaccine against melanoma was developed, showing strong luciferase activity and suppression of melanoma growth and metastasis in mice. The ternary complex mainly transfected into macrophages in the spleen, indicating its suitability for DNA vaccination. The ternary complex also exhibited low acute and liver toxicities in mice, confirming its safety profile.
