Targeting the Inside of Cells with Biologicals: Toxin Routes in a Therapeutic Context

Maximilian Ruschig et al.

Summary: Numerous toxins demonstrate routes for proteins to translocate from the extracellular space to the cytosol. Understanding these routes is important for therapeutic applications such as anti-toxin treatments, toxin-based drug delivery, and targeted delivery. While toxin routes may have lower delivery efficiencies, they can still be effective for certain applications, especially if the delivered cargoes have amplification effects or require only a small number of molecules. The mechanisms by which toxins enter cells may also affect their technological utility.

Full-coverage regulations of autophagy by ROS: from induction to maturation

Jing Zhou et al.

Summary: Autophagy is a well-conserved recycling process in response to stress, removing oxidized/damaged proteins and organelles to restrict further reactive oxygen species (ROS) production. Oxidative regulations occur throughout all stages of autophagy, including induction, nucleation, expansion, maturation, delivery and degradation, as well as gene transcription. Direct thiol oxidation of key proteins and certain chaperone-like proteins play roles in regulating different aspects of autophagy, while ROS can also inhibit autophagy through oxidation of key regulators.

A potential delivery system based on cholera toxin: A macromolecule carrier with multiple activities

Xianying He et al.

Summary: Macromolecular drug delivery systems, especially using Cholera toxin (CT) as a delivery system, show great potential in retaining drug activity and inducing specific immunity. However, the toxicity of native CT restricts its practical application, and thus, research on CT-based proteins with reduced virulence and enhanced adjuvant activity is necessary.

Targeted intracellular delivery of Cas13 and Cas9 nucleases using bacterial toxin-based platforms

Songhai Tian et al.

Summary: In this study, protein-based delivery systems were developed using detoxified single-chain bacterial toxins as carriers. These systems can deliver large protein cargoes into specific cells in a receptor-dependent manner. Delivery of Cas13a, CasRx, Cas9, and Cre recombinase had different effects on gene expression in various cell lines.

Jellyfish Bioprospecting in the Mediterranean Sea: Antioxidant and Lysozyme-Like Activities from Aurelia coerulea (Cnidaria, Scyphozoa) Extracts

Loredana Stabili et al.

Summary: The study focused on investigating the antioxidant and lysozyme antibacterial activities associated with extracts from different parts of A. coerulea. Extracts from the oral arms of A. coerulea showed significant antioxidant activity and the highest lysozyme-like activity, suggesting their potential as a source of novel cytolytic and cytotoxic products against marine bacteria. The wide geographic distribution and seasonal abundance of A. coerulea populations in coastal waters support the exploration of biotechnological applications of jellyfish biomasses in pharmaceutical, nutritional, and nutraceutical sectors.

Jellyfish venom proteins and their pharmacological potentials: A review

S. Amreen Nisa et al.

Summary: Recent research on marine invertebrates has shown that jellyfish venoms are rich in toxins used for hunting and defense, but can also have harmful effects on humans. The nematocyst, a complex intracellular system found in cnidarians, injects a venomous mixture into prey or predators. These venoms can induce cytotoxicity, ion channel blockade, membrane pore formation, cardiovascular failure, and lethal effects in laboratory animals.

Inflammatory Molecules Associated with Ultraviolet Radiation-Mediated Skin Aging

Tuba M. Ansary et al.

Summary: Skin is the largest and most complex organ in the human body, composed of different types of cells. Various environmental stressors, such as UVR, can accelerate skin aging by stimulating inflammatory molecules. UVR-induced skin aging is characterized by loss of elasticity, among other factors, and aged skin serves as a source of circulatory inflammatory molecules that further accelerate aging and contribute to aging-related diseases.

Cellular uptake of large biomolecules enabled by cell-surface-reactive cell-penetrating peptide additives

Anselm F. L. Schneider et al.

Summary: The challenge of delivering proteins into cells has been addressed by conjugating arginine-rich cell-penetrating peptides to the cell surface, enabling efficient delivery of protein-CPP conjugates. The use of thiol-reactive cell-penetrating peptides allows for simple co-incubation to deliver thiol-containing proteins, greatly simplifying protein and antibody delivery with minimal genetic or chemical engineering.

The Role of Thioredoxin/Peroxiredoxin in the beta-Cell Defense Against Oxidative Damage

Jennifer S. Stancill et al.

Summary: Oxidative stress is implicated in pancreatic beta-cell damage in type 1 and type 2 diabetes, potentially leading to dysfunction and death of beta cells. Beta cells are vulnerable to oxidative damage but have high expression of antioxidant enzymes which may help maintain their function and survival.
