Metallobiology of Lactobacillaceae in the gut microbiome

Uyen Huynh et al.

Summary: Lactobacillaceae are a diverse family of lactic acid bacteria that have beneficial effects on intestinal health, modulating the immune system and providing protection against pathogens. Metal ions, such as iron, zinc, and manganese, play a role in the microbial community composition and gastrointestinal diseases. However, the molecular mechanisms underlying the changes in metal availability and the response of Lactobacillaceae to metal limitation or excess are not well understood. This review provides an overview of these mechanisms and discusses the impact of iron, zinc, and manganese on Lactobacillaceae in the gut microbiota.

Recombinant Fusion Protein Vaccine Containing Clostridioides difficile FliC and FliD Protects Mice against C. difficile Infection

Shaohui Wang et al.

Summary: Bacterial flagella play crucial roles in infection by mediating host cell adhesion, invasion, auto-agglutination, colonization, biofilm formation, and the regulation and secretion of nonflagellar bacterial proteins associated with virulence. In this study, a fusion protein vaccine (FliCD) containing Clostridioides difficile flagellar proteins FliC and FliD was constructed. Immunization with FliCD induced robust IgG and IgA antibody responses, provided protection against C. difficile infection, and reduced C. difficile spore and toxin levels in the feces. Furthermore, the anti-FliCD serum inhibited the binding of C. difficile vegetative cells to HCT8 cells. These findings suggest that FliCD may serve as an effective vaccine candidate against CDI.

Nutritional immunity: the battle for nutrient metals at the host-pathogen interface

Caitlin C. Murdoch et al.

Summary: Trace metals play essential roles in the interactions between bacterial pathogens and animal hosts. They promote enzyme catalysis and stabilize reaction intermediates, while also preventing bacterial proliferation. The cell-specific and tissue-specific roles of distinct trace metals in shaping bacterial infections have implications for future research and new therapeutic development.

Improvement of the silver staining method for bacterial flagella

Jingsong Ye et al.

Summary: Flagella staining is a common method used in microbial research. In this study, we improved the Blendon staining method by adding two steps and compared the staining effects under four different culture conditions. The results showed that the staining effect was best in semi-solid culture medium and worst in liquid culture medium. This improved method is simple, low cost, and worthy of promotion.

Structural basis of Streptomyces transcription activation by zinc uptake regulator

Xu Yang et al.

Summary: The molecular mechanism of transcription activation by ScZur in Streptomyces coelicolor has been deciphered through cryo-EM structures and biochemical assays. ScZur binds to Zur-box sequences and interacts with Sc RNA polymerase and promoter DNA. These interactions are crucial for the transcription initiation of housekeeping genes in Streptomyces coelicolor.

Impairment of the Zn/Cd detoxification systems affects the ability of Salmonella to colonize Arabidopsis thaliana

Sabina Visconti et al.

Summary: The ability of Salmonella to colonize different environments depends on its response to fluctuations in micronutrient availability. Zinc, an essential micronutrient, plays a key role in bacterial physiology and can influence bacterial survival in plants. This study investigated the involvement of Salmonella's zinc homeostasis effectors in plant colonization, and found that Salmonella utilizes efflux pumps to export excess zinc and colonize plant tissues. The study also showed that metal availability in plant tissues affects bacterial persistence in the shoots.

Pectobacterium atrosepticum KDPG aldolase, Eda, participates in the Entner-Doudoroff pathway and independently inhibits expression of virulence determinants

Huan Wang et al.

Summary: The study demonstrates that the disruption of the Eda enzyme has a feedback effect on inhibiting pectin degradation and affects virulence and pectinase activity in Pectobacterium infection. Eda is shown to play an important role in building the arsenal of pectinases required during infection by Pectobacterium.

Arabidopsis bZIP19 and bZIP23 act as zinc sensors to control plant zinc status

Grmay H. Lilay et al.

Summary: Zinc (Zn) is an essential micronutrient for plants and animals, but how plants sense zinc is poorly understood. Two Arabidopsis bZIP transcription factors previously shown to modulate plant responses to zinc deficiency are now identified as zinc sensors in this study.
