Empagliflozin Ameliorates Ouabain-Induced Na+ and Ca2+ Dysregulations in Ventricular Myocytes in an Na+-Dependent Manner

Xiaodong Peng et al.

Summary: This study found that empagliflozin can improve Na+ and Ca2+ handling disorders induced by ouabain in a Na+-dependent manner. The research results suggest that empagliflozin exerts its therapeutic effects by inhibiting the Na+/H+ exchanger (NHE).

Regulation of Cardiac Contractility by the Alpha 2 Subunit of the Na+/K+-ATPase

Jonas Skogestad et al.

Summary: Cytosolic Na + concentrations play a crucial role in regulating cardiac excitation-contraction coupling and contractility. The inhibition of Na+/K+-ATPase activity can increase cardiac contractility by altering cytosolic Ca2+ levels. The alpha 2 isoform of NKA is considered the primary regulator of cardiac contractility, despite being less abundant than the alpha 1 isoform.

Summary: Cardiovascular diseases are the leading cause of death in industrialized countries, with hypoxia playing a central role in their pathogenesis. The review highlights the importance of HIFs and derived factors in the development and perpetuation of hypoxia in these diseases, categorizing them into acute and chronic hypoxic states to better understand the temporal relevance of HIFs in disease progression.

Summary: Diastolic dysfunction is a major feature of hypertrophic cardiomyopathy (HCM). Ischemic heart injury leads to HIF1-dependent differential splicing of CaMK2γ

Allison Lesher Williams et al.

Summary: Ischemic heart disease leading to heart failure is closely related to hypoxia inducible factor 1 (HIF1) playing a key role in the response to hypoxic injury. Studies have shown that inducible expression of an oxygen-stable form of HIF1 alpha in cardiomyocytes leads to rapid cardiac dilation and loss of contractility, potentially due to lower expression of excitation-contraction coupling genes and reduced calcium flux. Alternative splicing, an important mechanism in transcriptional regulation, was identified as playing a significant role in modifying gene expression in both the HIF-PPN hearts and post-myocardial infarction mice, indicating changes in cellular factors and metabolic pathways in pathological conditions.
