Sortilin regulates blood-brain barrier integrity

Andrea E. Toth et al.

Summary: Sortilin acts as a crucial regulatory protein for maintaining the tightness of the blood-brain barrier (BBB). Lack of sortilin results in BBB leakage, potentially due to alterations in signalling pathways, leading to compromised connections between brain endothelial cells.

Secreted Amyloid Precursor Protein Alpha, a Neuroprotective Protein in the Brain Has Widespread Effects on the Transcriptome and Proteome of Human Inducible Pluripotent Stem Cell-Derived Glutamatergic Neurons Related to Memory Mechanisms

Katie Peppercorn et al.

Summary: This study comprehensively analyzed the effects of secreted amyloid precursor protein alpha (sAPP alpha) on the transcriptome and proteome of human neurons. The findings showed that sAPP alpha has an impact on the molecular biology of learning and memory, consistent with previous studies on its memory enhancement and neuroprotective properties. The study also identified a set of differentially regulated proteins associated with Alzheimer's risk genes, APP processing, synaptogenesis, and other important cellular functions.

Hippocampal astrocytes encode reward location

Adi Doron et al.

Summary: This study used two-photon microscopy to investigate the real-time calcium activity of astrocytes in the hippocampus of awake mice. The researchers found that astrocytes exhibit persistent ramping activity towards the reward location in a familiar environment, but not in a new one. Shifting the reward location within a familiar environment also resulted in diminished ramping. After additional training, the ramping activity was re-established.

Aberrant astrocyte protein secretion contributes to altered neuronal development in multiple models of neurodevelopmental disorders

Alison L. M. Caldwell et al.

Summary: This study developed a cell culture system to investigate the impact of astrocyte protein secretion and gene expression changes in genetic neurodevelopmental disorders. It was found that astrocytes in these disorders increased the release of Igfbp2, which is an inhibitor of insulin-like growth factor (IGF). Blocking Igfbp2 partially rescued the inhibitory effects of Rett syndrome astrocytes, suggesting that increased astrocyte Igfbp2 contributes to decreased IGF signaling in neurodevelopmental disorders. The study identified increased BMP signaling as the upstream mechanism for protein secretion changes and blocking BMP signaling reversed inhibitory effects on neurite outgrowth. This work provides new targets for intervention in diverse neurodevelopmental disorders.

Lentivirus-Mediated Expression of Human Secreted Amyloid Precursor Protein-Alpha Promotes Long-Term Induction of Neuroprotective Genes and Pathways in a Mouse Model of Alzheimer's Disease

Margaret Ryan et al.

Summary: This study identified key molecular processes underlying the long-term neuroprotective and therapeutic effects of increasing sAPP alpha levels in vivo, including the regulation of GABA receptor signaling, cell survival, and inflammatory responses. Upstream gene regulatory analysis indicated sAPP alpha activation of Interleukin-4, which can counteract inflammatory changes in Alzheimer's disease.

Summary: The study revealed that miR-122-5p is selectively sorted and secreted into lung cancer EVs through binding to RNA-binding protein hnRNPA2B1, promoting the migration of liver cells. This may contribute to establishing a pre-metastatic micro-environment and hepatic metastasis of lung cancer.

Secreted Amyloid Precursor Protein-Alpha Enhances LTP Through the Synthesis and Trafficking of Ca2+-Permeable AMPA Receptors

Rhys W. Livingstone et al.

Summary: sAPP alpha enhances hippocampal LTP by promoting the cell-surface expression of GluA1 AMPARs and reducing GluA2 levels, affecting the localization and internalization of AMPARs at synapses, ultimately increasing synaptic levels of GluA1/2 AMPARs.

Astrocyte-Neuron Signaling in Synaptogenesis

Lili Shan et al.

Summary: Astrocytes play a crucial role in the central nervous system by dynamically communicating with neurons to regulate synaptic formation and maturation. Their heterogeneity allows them to boost synaptogenesis region-specifically. Future research may focus on utilizing organoids generated from human induced pluripotent stem cells to study the signaling pathways employed by astrocytes in synaptic development.
