Oxidized low-density lipoprotein associates with cardiovascular disease by a vicious cycle of atherosclerosis and inflammation: A systematic review and meta-analysis

Christin G. Hong et al.

Summary: This systematic review and meta-analysis explored the relationship between oxidized LDL and cardiovascular disease in patients with chronic inflammatory conditions. The findings indicate that oxidized LDL levels are significantly elevated in participants with cardiovascular disease in the presence of chronic inflammation. This suggests that oxidized LDL may serve as a useful biomarker for risk stratification of cardiovascular disease in patients with chronic inflammation.

Paraoxonase-1 (PON-1) Arylesterase Activity Levels in Patients with Coronary Artery Disease: A Meta-Analysis

Marco Zuin et al.

Summary: This study reviewed and compared the PON-1 arylesterase activity between coronary artery disease (CAD) and non-CAD patients. The results showed that CAD patients had significantly lower PON-1 arylesterase activity compared to controls, and CAD patients without diabetes mellitus (DM) had significantly higher PON-1 arylesterase activity.

A PON for All Seasons: Comparing Paraoxonase Enzyme Substrates, Activity and Action including the Role of PON3 in Health and Disease

Chrysan J. Mohammed et al.

Summary: Paraoxonases (PONs) are a family of hydrolytic enzymes that play an important role in various diseases, especially cardiovascular diseases. Among them, PON3 is the least studied member, but it has potential implications in diseases such as HIV and cancer. Identifying the specific substrates and activities of PONs is crucial for understanding their biological roles and biomedical applications.

B Vitamins Prevent Iron-Associated Brain Atrophy and Domain-Specific Effects of Iron, Copper, Aluminum, and Silicon on Cognition in Mild Cognitive Impairment

Hieronim Jakubowski et al.

Summary: The study highlights the effects of metals, silicon, and homocysteine on cognition and brain atrophy in MCI patients, and how B vitamin therapy can modify these relationships. Additionally, iron, copper, aluminum, and silicon were found to have domain-specific associations with cognition and were influenced by B vitamin therapy.

Summary: This study investigated the differences in PON1 and PON3 protein expression in the brain of a mouse model of AD, finding intense staining of these proteins in star-shaped cells surrounding A beta plaques. The results suggest that PON1 and PON3 may play a crucial role in preventing oxidative stress and lipid peroxidation in specific brain-cell types in AD pathology and potentially in other neurodegenerative diseases.

Paraoxonase 1 gene variants concerning cardiovascular mortality in conventional cigarette smokers and non-smokers treated with hemodialysis

Alicja E. Grzegorzewska et al.

Summary: This study investigated the association between different single nucleotide variants of the PON1 gene and cardiovascular mortality in hemodialysis patients, particularly in smokers. Specific genotypes and alleles were found to be associated with different types of deaths in smokers, non-smokers, diabetic, and non-diabetic individuals. These findings provide insights for identifying individuals at higher risk for cardiovascular mortality and the need for preventive measures.

Paraoxonase 1, B Vitamins Supplementation, and Mild Cognitive Impairment

Joanna Perla-Kajan et al.

Summary: The study found that PON1 status is associated with cognitive function in individuals with mild cognitive impairment (MCI), with PhAcase activity showing significant correlations with global cognition, verbal episodic memory, and attention/processing speed. In addition to PON1, baseline levels of iron and triglycerides, fatty acids, anti-N-Hcy-protein autoantibodies, and BDNF V66M genotype were also related to cognitive function in MCI patients. B-vitamins were able to mitigate the associations of PON1 and other variables with cognition.

Changes in redox plasma proteome of Pon1-/- mice are exacerbated by a hyperhomocysteinemic diet

Summary: The inactivation of the Pon1 gene resulted in dysregulation of redox-related proteins in mice, with a higher impact observed in mice fed with a hyper-homocysteinemic diet. This dysregulation of key proteins involved in antioxidant defenses and other biological processes provides insight into the pro-oxidant and proatherogenic phenotypes seen in Pon1(-/-) mice and humans.
