Factors associated with subjective knee joint stiffness in people with knee osteoarthritis: A systematic review

Daisuke Uritani et al.

Summary: This study aimed to explore the factors associated with subjective knee stiffness in patients with knee osteoarthritis. The results showed that worse preoperative subjective knee stiffness was associated with worse pain, subjective knee stiffness, and patient satisfaction at 1 year after total knee arthroplasty. In addition, worse subjective knee stiffness was associated with future degenerative changes in the knee joint.

Mesenchymal stromal cell-based therapy for cartilage regeneration in knee osteoarthritis

Xiao-Na Xiang et al.

Summary: Osteoarthritis is a common degenerative disease that causes high socioeconomic costs and disability rates. The knee joint is most commonly affected, characterized by progressive destruction of articular cartilage, loss of extracellular matrix, and inflammation. MSC-based therapy has been explored as a new regenerative treatment for knee osteoarthritis. However, the detailed functions and mechanisms of MSC-based therapy, especially for cartilage regeneration, have not been explained.

Hypoxia Induces DPSC Differentiation versus a Neurogenic Phenotype by the Paracrine Mechanism

Simona Delle Monache et al.

Summary: Studies suggest that hypoxia treatment of DPSCs promotes their differentiation into neuronal cells in a time-dependent manner. Additionally, the conditioned media from hypoxia-stimulated DPSCs can induce neural differentiation in other neural cells.

Activation of TRPV4 by mechanical, osmotic or pharmaceutical stimulation is anti-inflammatory blocking IL-1 beta mediated articular cartilage matrix destruction

S. Fu et al.

Summary: The study demonstrates that TRPV4 plays an anti-inflammatory role in chondrocytes and cartilage explants, regulating tubulin modifications and cilia length to suppress interleukin-1β-mediated inflammatory signaling.

Dental Pulp-Derived Mesenchymal Stem Cells for Modeling Genetic Disorders

Keiji Masuda et al.

Summary: Dental pulp stem cells, derived from multipotent neural crest cells, have high proliferative capacity and multipotent potential for various applications. Stem cells donated by patients with genetic disorders can serve as disease models. Ethical considerations and potential applications of these stem cells donated and reused are worth exploring.

A dysfunctional TRPV4-GSK3β pathway prevents osteoarthritic chondrocytes from sensing changes in extracellular matrix viscoelasticity

Pranay Agarwal et al.

Summary: The viscoelasticity of alginate hydrogels regulates cell volume in healthy chondrocytes but not in osteoarthritic chondrocytes. This difference in response is associated with changes in anabolic gene expression, secretion of pro-inflammatory cytokines, and modulation of intracellular calcium. Dysfunctional TRPV4-GSK3 beta pathway in osteoarthritic chondrocytes renders them unable to respond to viscoelastic changes in the extracellular matrix.

Regenerative Potential of DPSCs and Revascularization: Direct, Paracrine or Autocrine Effect?

Vincenzo Mattei et al.

Summary: The newly discovered dental pulp derived stem cells (DPSCs) have the potential to be tools for regenerative medicine, originating from the neural crest and involved in dentin homeostasis. They contribute to bone remodeling and differentiation into various tissues, and also influence angiogenesis by releasing secretory factors or differentiating into vascular and perivascular cells.
