Identification of a candidate gene underlying qHKW3, a QTL for hundred-kernel weight in maize

Summary: Kernel size and weight are important traits for maize yield. In this study, we developed a chromosome segment substitution line with smaller kernel size and lower kernel weight. Histological analysis revealed that the slower kernel filling rate contributed to the small-kernel size and reduced hundred-kernel weight in this line. We identified a quantitative trait locus (QTL), qHKW3, which explained 23% of the phenotypic variation in hundred-kernel weight. By fine mapping, we identified the gene Zm00001d044081 as the causal gene for qHKW3, encoding the homeobox-leucine zipper protein ATHB-4. The expression level of Zm00001d044081 showed significant differences between the near-isogenic lines, and our association mapping analysis confirmed its effects on kernel size and weight in a larger panel. This gene represents a promising target for genetic improvement of maize yield.

OsBSK2, a putative brassinosteroid-signalling kinase, positively controls grain size in rice

Summary: Grain size is an important trait in rice that affects grain yield. This study identified a mutant, glw10, which had reduced grain length and weight. The gene GLW10 was found to encode a putative brassinosteroid signaling kinase, OsBSK2, which regulates cell expansion and grain size. OsBSK2 interacts with the BR receptor kinase OsBRI1 and can form homodimers or heterodimers with OsBSK3 and OsBSK4. The OsBSKs are highly expressed in young panicles and spikelet hulls, indicating their role in controlling grain size. This study provides new insights into the genetic mechanisms of grain size regulation in rice.

The OsEIL1-OsERF115-target gene regulatory module controls grain size and weight in rice

Summary: The ethylene response factor OsERF115 functions as a key downstream regulator for ethylene-mediated grain development, positively regulating grain size and weight. OsERF115 is strongly induced by ethylene and acts as a transcriptional repressor to modulate a set of grain-size genes. Natural variations in the OsERF115 promoter contribute to grain-size diversity. OsERF115 orthologues are only found in grass species, indicating a conserved and unique role in grain development of cereal crops.

KNOX II transcription factor HOS59 functions in regulating rice grain size

Summary: In this study, the genome-wide binding sites of HOS59, a member of the rice KNOX Class II subfamily, were characterized using ChIP-seq technology. Trait Ontology analysis identified 9705 downstream target genes related to plant morphology and stress traits. Validation experiments confirmed the role of HOS59 as a repressor of these target genes, negatively regulating rice grain size and plant architecture. The identification of downstream target genes and interaction proteins of HOS59 improves our understanding of the KNOX regulatory networks.
