Toward Overcoming Pyrethroid Resistance in Mosquito Control: The Role of Sodium Channel Blocker Insecticides

Impact of insecticide resistance on malaria vector competence: a literature review

Summary: Since its first report in Anopheles mosquitoes in 1950s, insecticide resistance has rapidly spread to most sub-Saharan African malaria-endemic countries, endangering the success of vector control efforts and leading to a resurgence of malaria cases. Supported by four main mechanisms, this phenomenon is associated with intrinsic changes in resistant insect vectors that may affect the development of invading Plasmodium parasites. A literature review was conducted to evaluate the impact of insecticide resistance and associated mechanisms on key determinants of malaria vector competence, revealing contradictory evidence on the permissiveness of insecticide-resistant vectors to Plasmodium infections. The current understanding of factors such as immunity and microbiota communities in insecticide-resistant vectors is insufficient to definitively assess the epidemiological importance of these vectors compared to susceptible counterparts, calling for further studies to fill knowledge gaps in predicting malaria epidemiology amidst the ongoing selection and spread of insecticide-resistant vectors.

Characterizing fenestration size in sodium channel subtypes and their accessibility to inhibitors

Summary: Voltage-gated sodium channels play a crucial role in the electrical activity of nerve and muscle cells, with humans having nine different subtypes. Research indicates that there are four lateral fenestrations within these channels influencing the access of pore blockers. Although there are structural differences among the subtypes, a common pattern in the dimensions of the fenestrations has been observed, with one particular fenestration appearing most favorable for drug access in all subtypes. These findings are significant for the development of pharmacological treatments and the analysis of disease mutations in sodium channel subtypes.
