mtUPR Modulation as a Therapeutic Target for Primary and Secondary Mitochondrial Diseases

Paula Cilleros-Holgado et al.

Summary: Mitochondrial dysfunction plays a crucial role in various diseases, but effective treatments for mitochondrial diseases are still lacking. Therefore, researchers are exploring new therapeutic approaches, such as modulating the mitochondrial unfolded protein response (mtUPR). Activation of mtUPR can promote cell homeostasis and improve lifespan and disease conditions in biological models of mitochondrial dysfunction. However, mtUPR activation may also promote tumor progression and lead to undesirable side effects, such as increased heteroplasmy of mitochondrial DNA mutations.

Impairment of mitophagy and autophagy accompanies calcific aortic valve stenosis favouring cell death and the severity of disease

Giampaolo Morciano et al.

Summary: This observational study aims to establish the trend of autophagy in CAVS patients, analyze the involvement of other determinants impacting mitochondrial quality control mechanisms, and explore potential pharmacological interventions. The results show deficiencies in mitochondrial respiration, ATP production, and alterations in biogenesis and mitophagy pathways in the calcified valves, suggesting a rapamycin-based therapeutic strategy to revert pathological phenotypes. Boosting autophagy and mitophagy could potentially counteract cell death and sustain cell functions in the long term.

PTP1B Inhibition Improves Mitochondria Dynamics to Alleviate Calcific Aortic Valve Disease Via Regulating OPA1 Homeostasis

Feng Liu et al.

Summary: This study evaluated the role of protein tyrosine phosphatase 1B (PTP1B) inhibition in calcific aortic valve disease (CAVD). The findings showed that up-regulation of PTP1B was involved in calcification of the aortic valve and PTP1B inhibition had beneficial effects in preventing fibrocalcific response. Additionally, PTP1B was found to regulate mitochondrial homeostasis in valvular interstitial cell osteogenesis. These findings suggest that PTP1B could be a potential target for preventing aortic valve calcification in patients with CAVD.

Aorta smooth muscle-on-a-chip reveals impaired mitochondrial dynamics as a therapeutic target for aortic aneurysm in bicuspid aortic valve disease

Mieradilijiang Abudupataer et al.

Summary: Bicuspid aortic valve (BAV) is a common congenital cardiovascular disease associated with thoracic aortic aneurysm (TAA). Insufficiency of NOTCH1 expression is related to BAV-TAA, with impaired mitochondrial dynamics observed. Knockdown of NOTCH1 in HAoSMCs resulted in reduced contractile phenotype and attenuated mitochondrial fusion, which could be partially rescued by mitochondrial fusion activators or fission inhibitor. This study suggests impaired mitochondrial dynamics as a potential therapeutic target for BAV-TAA.
