Trabectedin impairs invasiveness and metastasis in adrenocortical carcinoma preclinical models

Andrea Abate et al.

Summary: Trabectedin can inhibit the invasiveness and metastasis formation of adrenocortical carcinoma (ACC) cells, reducing the tumor size and decreasing the rate of embryo metastasis. These results provide the rationale for a clinical evaluation of the efficacy of trabectedin in ACC patients.

Ribociclib Cytotoxicity Alone or Combined With Progesterone and/or Mitotane in in Vitro Adrenocortical Carcinoma Cells

Andrea Abate et al.

Summary: Ribociclib might induce apoptotic cell death and reduce cell proliferation rate through interfering with cell-cycle progression. The effect on cell viability is enhanced when combined with progesterone and/or mitotane.

A Comprehensive Investigation of Steroidogenic Signaling in Classical and New Experimental Cell Models of Adrenocortical Carcinoma

Sandra Sigala et al.

Summary: In this study, the genetic and pathophysiological characteristics of three specific adrenocortical carcinoma cell lines were analyzed, revealing their importance in reflecting the heterogeneities of ACC patients. This provides a more accurate model for preclinical studies.

Advances in adrenocortical carcinoma pharmacotherapy: what is the current state of the art?

Valentina Cremaschi et al.

Summary: The ADIUVO trial showed that adjuvant mitotane is not beneficial for patients with a good prognosis. Future strategies aim to strengthen adjuvant therapy for patients at high risk of relapse. While modern targeted therapies have shown significant cytotoxicity in preclinical studies for advanced/metastatic ACC, results from studies in patients have been disappointing so far.

Modulation of Autophagy in Adrenal Tumors

Diana Sousa et al.

Summary: Adrenal masses are common in humans, with most being benign and non-functioning. However, rare malignant tumors have poor prognosis due to the absence of effective treatments. Autophagy, a cellular process involved in the degradation of organelles and proteins, has been targeted by chemotherapy drugs. The role of autophagy in cancer is paradoxical, with studies showing both pro-survival and cell death effects. In adrenal tumors, autophagy has been studied using cell lines, but clinical studies are still needed to understand its role.

An update on adrenocortical cell lines of human origin

Sandra Sigala et al.

Summary: This review provides an overview of the management and treatment progress of adrenocortical carcinoma. Complete surgical resection is the only potentially curative option, but systemic treatments are needed for advanced cases. The lack of preclinical models reflecting patient heterogeneities and drug-resistant phenotypes has hindered the development of new therapies.

Caffeine Inhibits Direct and Indirect Angiogenesis in Zebrafish Embryos

Ram Manohar Basnet et al.

Summary: The study showed that caffeine treatment results in a shortening and thinning of blood vessels in zebrafish embryos, along with decreased expression of vascular marker genes and a reduction in the number of interconnecting vessels. Caffeine is also able to block angiogenesis induced by exogenous Fibroblast Growth Factor 2 or cells producing Fibroblast Growth Factor 2. Overall, the results indicate an inhibitory effect of caffeine on both direct and indirect angiogenesis.

Progestins as Anticancer Drugs and Chemosensitizers, New Targets and Applications

Tatiana A. Fedotcheva et al.

Summary: Progesterone and progestins play crucial roles in regulating cell differentiation, proliferation, and cell cycle progression, and have been widely used in contraception, pregnancy maintenance, and hormone replacement therapy. Recent studies have demonstrated their effectiveness in treating hormone-sensitive tumors, mainly through their cytotoxic and chemosensitizing effects on cancer cells. The therapeutic potential of progestins lies in their ability to modulate key signaling pathways, inhibit survival mechanisms, and enhance chemosensitivity in hormone-dependent tumors.

Novel Insights into the Molecular Regulation of Ribonucleotide Reductase in Adrenocortical Carcinoma Treatment

Christina Bothou et al.

Summary: The current clinical treatment options for adrenocortical carcinomas (ACCs) are unsatisfactory, and novel therapies have shown promise in preclinical studies but disappointing clinical translation. The combination of gemcitabine and cisplatin has shown promising anti-tumor efficacy against ACC cell lines, but resistance mechanisms have been identified. Modulation of ribonucleotide reductase (RNR)-related pathways may offer a new therapeutic option for ACC.
