Redox-dependent internalization of the purinergic P2Y6 receptor limits colitis progression

Kazuhiro Nishiyama et al.

Summary: This study identified a mechanism called redox-dependent alternative internalization (REDAI) that promotes the internalization and degradation of the purinergic P2Y(6) receptor (P2Y(6)R). The researchers found that both synthetic and natural compounds containing electrophilic isothiocyanate groups can modify P2Y(6)R and facilitate its internalization and degradation. This mechanism, mediated by cysteine modification, may also play a role in the internalization of other G protein-coupled receptors (GPCRs) and potentially limit the progression of colitis.

Inhibition of P2Y6 receptor expression in Kupffer cells alleviates alcoholic steatohepatitis in mice

Fei Yuan et al.

Summary: In this study, the role of P2Y6 in alcoholic steatohepatitis was investigated. P2Y6 levels were found to be elevated in Kupffer cells in mice with alcoholic steatohepatitis and ethanol-induced RAW264.7 cells. Inhibition of P2Y6 showed improvement in steatosis and inflammation, while P2Y6 activation had contrasting results. Furthermore, P2Y6 was found to regulate the activation of the p38 MAPK signaling pathway through mediating calcium influx. Targeting P2Y6 signaling may be a potentially effective strategy for the treatment of alcoholic steatohepatitis.

Heterogeneity of non-alcoholic fatty liver disease (NAFLD): Implication for cardiovascular risk stratification

Francesco Baratta et al.

Summary: NAFLD is the most common liver disease worldwide and is strongly linked to atherosclerotic cardiovascular disease. It is a heterogeneous condition with various sub-phenotypes and outcomes, ranging from cardiovascular complications to liver damage progression. The contribution of environmental, metabolic, and genetic factors underlies this heterogeneity.

Gut bacteria alleviate smoking-related NASH by degrading gut nicotine

Bo Chen et al.

Summary: Tobacco smoking is positively correlated with non-alcoholic fatty liver disease (NAFLD), but the underlying mechanism is unclear. This study found that nicotine accumulates in the intestine during tobacco smoking and activates intestinal AMPK alpha. The gut bacterium Bacteroides xylanisolvens is identified as an effective nicotine degrader. These findings highlight the role of intestinal nicotine accumulation in NAFLD progression and the presence of an endogenous bacterium in the human intestine that can metabolize nicotine.

The Purinergic P2X7 Receptor-NLRP3 Inflammasome Pathway: A New Target in Alcoholic Liver Disease?

Brendan Le Dare et al.

Summary: Alcohol consumption is closely associated with the development and progression of many non-communicable inflammatory diseases, particularly in the liver. Recent research suggests that Kupffer cells play a key role in the pathophysiology of alcoholic liver disease. Therefore, modulating purinergic signaling may represent a new approach for treating alcoholic liver disease.

Summary: The study shows that deficiency of P2Y2R can improve NAFLD induced by high-fat diet through various pathways, including enhancing insulin resistance, inhibiting hepatic lipid accumulation and injury, reducing fatty acid synthesis, and increasing fatty acid oxidation. This suggests that P2Y2R may be a promising therapeutic target for NAFLD.

Summary: Advances in molecular profiling have enabled the creation of high-resolution single-cell type maps of human tissues, combining single-cell transcriptomics analysis with spatial antibody-based protein profiling. This technology allows researchers to explore the expression of human protein-coding genes and determine the number of elevated genes in different cell types, facilitating comparisons with bulk transcriptomics data. The analysis reveals distinct expression clusters corresponding to cell types with similar functions within and between organs.
