Does Metformin Treatment in Pediatric Population Cause Vitamin B12 Deficiency?

Ozen Tas et al.

Summary: This prospective study aimed to evaluate the effects of metformin on blood vitamin B12 levels in children and adolescents. The results showed no significant changes in serum vitamin B12, homocysteine, MMA, or holo-TC-II levels with metformin therapy at 6-month and 12-month follow-ups. Long-term prospective studies with high-dose metformin treatment in the pediatric population are needed to confirm these results.

Summary: This study examined the changes in vitamin B12 levels in pediatric patients being treated with metformin. The results showed that there was no significant decrease in mean vitamin B12 levels in patients treated with metformin, except for those taking high doses with good compliance. The overall vitamin B12 levels remained within the normal reference range.

Summary: Obesity has negative impacts on the health and well-being of children and their families, leading to various acute problems. Preventive measures such as healthy diet, physical activity, adequate sleep and limited screen time should be implemented. Pharmacological management is limited, and surgery is an effective option.

Prevalence of multiple non-communicable diseases risk factors among adolescents in 140 countries: A population-based study

Tuhin Biswas et al.

Summary: This study assessed the global status of clustering of common modifiable non-communicable disease (NCD) risk factors among adolescents. The findings indicate a high prevalence of four or more NCD risk factors in adolescents globally, with variations between countries. Insufficient vegetable consumption, insufficient fruit consumption, and physical inactivity were identified as the most prevalent risk factors in all regions.

Summary: Metformin is the first-line oral glucose-lowering drug for type 2 diabetes, but long-term use may lead to vitamin B12 deficiency. There is currently no definite guideline for screening vitamin B12 deficiency in patients on metformin therapy.

Association of MTHFR (rs 1801133) gene polymorphism with biochemical markers of B12 deficiency in type 2 diabetes mellitus patients on metformin therapy

Ananda Vayaravel Cassinadane et al.

Summary: The study investigated the association between MTHFR gene polymorphism and metformin-induced vitamin B12 deficiency in T2DM patients, finding that the T allele in MTHFR gene may increase the risk of vitamin B12 deficiency in T2DM patients on metformin therapy. Additionally, the TT genotype of MTHFR gene showed a high likelihood ratio in predicting folic acid, MMA, and Hcy levels in T2DM patients on metformin therapy.

Can Metformin Exert as an Active Drug on Endothelial Dysfunction in Diabetic Subjects?

Teresa Salvatore et al.

Summary: Cardiovascular mortality is a major cause of death in patients with type 2 diabetes, with endothelial dysfunction being a significant risk factor. Metformin, a milestone in T2DM treatment, has been shown to have beneficial effects on endothelial dysfunction by inhibiting inflammation and improving endothelial function through AMPK activation.

Summary: Metformin exposure during pregnancy can lead to increased risk of small for gestational age births and childhood obesity in offspring, potentially through effects on maternal vitamin B12 deficiency and folate imbalance. Further research is needed to elucidate the mechanisms by which metformin impacts fetal growth and long-term health outcomes.
