Summary: Endocrine disrupting chemicals (EDCs) can have adverse effects on women's reproductive health. Current screening methods lack tests to assess the effects of EDCs on oocyte maturation and embryo development. This study describes a screening method using bovine models to evaluate the impact of EDCs on oocyte maturation and early embryo development.

Genome-wide identification of enhancers and transcription factors regulating the myogenic differentiation of bovine satellite cells

Pengcheng Lyu et al.

Summary: Tens of thousands of H3K27ac-marked active enhancers have been identified from bovine satellite cells before or during differentiation. These enhancers contain binding sites not only for transcription factors whose role in satellite cell differentiation is well known but also for transcription factors whose role in satellite cell differentiation is unknown. These enhancers and transcription factors are valuable resources for understanding the complex mechanism that mediates gene expression during satellite cell differentiation.

Summary: Culturing porcine oocytes in a medium containing FGF2, LIF and IGF1 (FLI) significantly enhances their developmental potential. Analyzing the time course of MAPK3/1 and AKT activation, as well as gene expression controlled by these kinases, suggests that both MAPK3/1 and AKT play crucial roles in improving oocyte quality in FLI medium.

RUNX1 Regulates a Transcription Program That Affects the Dynamics of Cell Cycle Entry of Naive Resting B Cells

Inesa Thomsen et al.

Summary: RUNX1 regulates a gene expression program in mouse B cells, affecting cell cycle dynamics in response to BCR stimulation. It interacts with SRCAP to recruit it to the promoter and enhancer regions of the Ccnd2 gene. Knockout of Runx1 leads to increased binding of BRG1 at the Ccnd2 and Rbpj promoters after BCR stimulation, resulting in hyperresponsiveness of B cells and potential involvement in regulating B cell tolerance.

SMAD4 Feedback Activates the Canonical TGF-β Family Signaling Pathways

Lu Liu et al.

Summary: The study illustrates a novel mechanism in which SMAD4, as the only Co-SMAD, positively regulates the canonical TGF-beta family signaling pathways in granulosa cells (GCs) by interacting with the core promoters of upstream receptors through the recruitment of three coactivators (c-JUN, CREB1, and SP1). Different interaction modes between SMAD4 and coactivators were identified through reciprocal ChIP assays, expanding the understanding of the feedback regulation modes of TGF-beta family signaling pathways in ovarian GCs.

TMEM182 interacts with integrin beta 1 and regulates myoblast differentiation and muscle regeneration

Wen Luo et al.

Summary: TMEM182 is specifically expressed in skeletal muscle and adipose tissue, regulated transcriptionally by MyoD1. Functionally, TMEM182 inhibits myoblast differentiation and fusion. In vivo studies show that TMEM182 induces muscle fibre atrophy and delays muscle regeneration, while TMEM182 knockout results in increased body weight, muscle mass, and accelerated skeletal muscle regeneration. Moreover, TMEM182 interacts with ITGB1 to modulate ITGB1 activation and downstream signaling pathways during myogenesis.
