Defective mitophagy in aged macrophages promotes mitochondrial DNA cytosolic leakage to activate STING signaling during liver sterile inflammation

Weizhe Zhong et al.

Summary: Aging impairs mitophagy and lysosomal function in macrophages, leading to the cytosolic release of mtDNA and increased STING activation. This promotes sterile inflammatory liver injury in aged patients. The findings suggest potential therapeutic targets for treating aged patients with sterile inflammatory liver injury.

The interaction between STING and NCOA4 exacerbates lethal sepsis by orchestrating ferroptosis and inflammatory responses in macrophages

Jie Wu et al.

Summary: The mechanism by which STING causes lethal sepsis through inducing macrophage ferroptosis in an interferon-independent manner is explored. The interaction between STING and NCOA4 is found to be critical in this process. The selective 20-HETE synthase inhibitor, HET0016, is shown to reduce STING-induced ferroptosis and mortality in septic mice. These findings provide insights into sepsis and potential therapeutic targets.

STING signaling sensing of DRP1-dependent mtDNA release in kupffer cells contributes to lipopolysaccharide-induced liver injury in mice

Qin Zhang et al.

Summary: This study demonstrated the important role of STING signaling in septic liver injury, with significant effects on liver function protection and attenuation of systemic inflammatory response. In addition, STING signaling activation resulted in LPS-induced liver injury through promoting hepatocyte death. Release of mitochondrial DNA (mtDNA) was identified as one of the mechanisms for STING signaling activation.

Astrocytic IGF-1 and IGF-1R Orchestrate Mitophagy in Traumatic Brain Injury via Exosomal miR-let-7e

Ren Dabin et al.

Summary: Defective brain hormonal signaling and autophagy are associated with neurodegeneration after brain insults. This study demonstrates that reduced plasma IGF-1 is correlated with cognitive impairment in traumatic brain injury (TBI) patients. Overexpression of astrocytic IGF-1 improves cognitive dysfunction and mitophagy in a TBI mouse model. Mechanistically, IGF-1 transcriptionally regulates decapping mRNA2 (Dcp2) and miR-let-7, along with IGF-1R, to orchestrate mitophagy in TBI.

Exosomal miR-21 secreted by IL-1β-primed-mesenchymal stem cells induces macrophage M2 polarization and ameliorates sepsis

Mengying Yao et al.

Summary: This study characterized the effects of miR-21 delivered by exosomes from MSCs on macrophage polarization in a mouse sepsis model. Beta MSC-derived exosomes induced M2-like polarization of macrophages more effectively, improving survival rates in septic mice. Exosomal miR-21 was identified as a key mediator of the immunomodulatory properties of MSCs induced by IL-1 beta pretreatment, providing a potential therapeutic basis for sepsis.

Exosomes from TNF-α-treated human gingiva-derived MSCs enhance M2 macrophage polarization and inhibit periodontal bone loss

Yuki Nakao et al.

Summary: Preconditioning gingival tissue-derived MSCs (GMSCs) with tumor necrosis factor-alpha (TNF-alpha) enhances the secretion of exosomes and the expression of CD73, promoting antiinflammatory M2 macrophage polarization. GMSC-derived exosomes exhibit significant ability to regulate inflammation and osteoclastogenesis, providing a potential therapeutic approach for periodontitis. The findings suggest a promising strategy for treating inflammatory osteoimmune disorders using TNF-alpha-preconditioned GMSC-derived exosomes.

TNF leads to mtDNA release and cGAS/STING-dependent interferon responses that support inflammatory arthritis

Joschka Willemsen et al.

Summary: The study elucidates a molecular mechanism linking TNF to type-I interferon signaling, demonstrating that TNF triggers a cGAS-STING-dependent interferon response and inhibits PINK1-mediated mitophagy. This leads to altered mitochondrial function, increased cytosolic mtDNA levels, and a transcriptional response that mimics macrophages from rheumatoid arthritis patients. Additionally, in an inflammatory arthritis mouse model, cGAS deficiency blocked interferon responses and reduced inflammatory cell infiltration and joint swelling. Therapeutic targeting of cGAS/STING in TNF-driven diseases may offer potential benefits.
