Systemic Interleukins' Profile in Early and Advanced Colorectal Cancer

Paulina Czajka-Francuz et al.

Summary: Tumor microenvironment (TME) consists of tumor, stromal, and immune cells interacting with each other. Early and advanced colorectal tumors have different structures and altered levels of serum cytokines. The crosstalk among TME infiltrating cells can influence patients' prognosis by shifting the balance towards immune suppression or pro-inflammatory, antitumor responses. Understanding TME processes could aid in therapeutic planning and accurate patient prognosis.

Key Players of the Immunosuppressive Tumor Microenvironment and Emerging Therapeutic Strategies

Kevin Park et al.

Summary: The tumor microenvironment is a battlefield for immune cells and tumor cells, and immune checkpoint inhibitors have shown promise in cancer treatment. However, response rates are low and resistance is common. Understanding the tumor microenvironment is crucial for developing strategies to overcome resistance, and ongoing clinical trials are investigating combination treatments targeting specific tumor and microenvironment characteristics.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Resident memory T cells in tumor-distant tissues fortify against metastasis formation

Laura S. Christian et al.

Summary: Using a murine breast cancer model, researchers found that resident memory T cells (TRMs) develop in the tumor, contralateral mammary mucosa, and pre-metastatic lung. Single-cell RNA sequencing of TRMs revealed two phenotypically distinct populations, sharing TCR clonotypes and transcriptomes with intratumoral effector/effector memory T cells. Releasing CXCR6 retention in the primary tumor can amplify tumor-derived TRMs in the lung and lead to superior protection against metastases in a potential strategy to prevent metastasis in clinical oncology.

Immunosuppressive Microenvironment Revealed by Immune Cell Landscape in Pre-metastatic Liver of Colorectal Cancer

Dongqiang Zeng et al.

Summary: This study elucidated the alterations of MDSCs during pre-metastatic niche transformation, highlighting their important role in immune cell infiltration. High frequencies of MDSCs in the pre-metastatic liver contribute to the secretion of immunosuppressive factors, leading to sustained immune suppression. Additionally, metabolic reprogramming and chemokines were identified as potential mechanisms for MDSC recruitment in the pre-metastatic liver.

Dynamic stromal cellular reaction throughout human colorectal adenoma-carcinoma sequence: A role of TH17/IL-17A

Guanglin Cui et al.

Summary: Accumulating data show that the tumor stroma undergoes rapid changes to create a supportive environment for disease progression. The activation of TH17/IL-17A network is significantly associated with dynamic stromal cellular response throughout the adenoma-carcinoma sequence, potentially promoting colorectal cancer initiation and progression.
