Enhancing the analytical performance of paper lateral fl ow assays: From chemistry to engineering

Summary: Paper-based lateral flow assays (LFAs) have achieved significant success in point-of-care diagnostics, however, they still face challenges in sensitivity, specificity, selectivity, reproducibility, and multiplex capability. Potential solutions in chemistry, nanobiotechnology, and engineering can be applied to enhance sensitivity, improve specificity, optimize selectivity, and develop better controls for reproducibility, ultimately paving the way for more efficient LFAs in various fields such as clinical settings, food safety, environment monitoring, and biosecurity.

Ultrasensitive and Highly Specific Lateral Flow Assays for Point-of-Care Diagnosis

Summary: LFAs, paper-based point-of-care diagnostic tools, have traditionally lower sensitivities and specificities than standard laboratory tests. Recent efforts have focused on optimizing assay kinetics, signal amplification, and sample preamplification to increase sensitivities, as well as improving specificity through high-affinity molecules and assay optimization. With continuing improvements, LFAs may soon offer performance competitive with laboratory techniques while retaining a rapid format.

Evaporation-Induced Diffusion Acceleration in Liquid-Filled Porous Materials

Summary: It was found that evaporation can significantly accelerate diffusion rates and alter concentration distributions in liquid-filled porous materials. A convective diffusion model was proposed based on the evaporation-induced flow and diffusion mechanism, which was experimentally validated. The study showed that evaporation-induced diffusion overwhelms molecular diffusion even at high relative humidities, with flow velocities comparable to those found in biological and biomedical systems.
