SH2 Domain-Containing Phosphatase 2 Inhibition Attenuates Osteoarthritis by Maintaining Homeostasis of Cartilage Metabolism via the Docking Protein 1/Uridine Phosphorylase 1/Uridine Cascade

Qianqian Liu et al.

Summary: In this study, the researchers identified SHP-2 as a novel regulator of cartilage homeostasis in osteoarthritis (OA). Inhibition of SHP-2 improved OA progression by maintaining the balance of cartilage metabolism.

Increased levels of sirtuin-1 in systemic lupus erythematosus

Chan Yang et al.

Summary: This study investigated the levels of plasma SIRT1 in systemic lupus erythematosus (SLE) patients and discussed its potential as a biomarker for SLE. The results showed that SLE patients had elevated levels of SIRT1 in plasma compared to healthy controls, and the concentration of plasma SIRT1 was significantly associated with disease activity. The analysis of receiver operating characteristic (ROC) curve demonstrated that plasma SIRT1 had a good ability to distinguish SLE from other rheumatic diseases and healthy controls.

SHP2 allosteric inhibitor TK-453 alleviates psoriasis-like skin inflammation in mice via inhibition of IL-23/Th17 axis

Meijing Wang et al.

Summary: This study identified TK-453 as a lead compound with strong affinity for SHP2, demonstrating its potential as a therapeutic target for psoriasis. TK-453 significantly improved skin inflammation in mice by inhibiting the IL-23/Th17 axis.

Characteristics and risk factors of severe coronary artery disease in systemic lupus erythematosus: A multicenter, Chinese Rheumatism Date Center database study

Weiping Ci et al.

Summary: This study aimed to assess the characteristics and risk factors of severe coronary artery disease (CAD) in patients with systemic lupus erythematosus (SLE). The results showed that age, major CAD risk factors, SLE Disease Activity Index 2000 (SLEDAI-2K), hyperuricemia, corticosteroid exposure, and positive antiphospholipid antibody (APL) were independent risk factors for severe CAD in SLE patients.

Therapeutic effect of the injectable thermosensitive hydrogel loaded with SHP099 on intervertebral disc degeneration

Jingcheng Wang et al.

Summary: The study revealed that SHP2 is highly expressed in degenerative IDD, and its overexpression inhibits the expression of Sox9 in NP cells, leading to decreased expression of key proteins and IDD development. SHP099 was able to reverse the degeneration of NP cells in vitro and showed therapeutic effects on IDD in rats when administered through injectable thermosensitive hydrogels.
