STAT3 and SPI1, may lead to the immune system dysregulation and heterotopic ossification in ankylosing spondylitis

Tuo Liang et al.

Summary: This study aimed to identify biomarkers for diagnosis and reveal immune microenvironment changes in ankylosing spondylitis (AS). The results indicate that STAT3 and SPI1 may be key cytokine receptors involved in disease progression in AS, with upregulated NKT cells and downregulated Th1 cells in AS patients.

CXCL12/CXCR4-Rac1-mediated migration of osteogenic precursor cells contributes to pathological new bone formation in ankylosing spondylitis

Haowen Cui et al.

Summary: This study identified CXCL12 as a critical contributor to pathological new bone formation in ankylosing spondylitis (AS) through recruitment of osteogenic precursor cells (OPCs). Inhibition of the CXCL12/CXCR4 axis attenuated OPCs migration and subsequent pathological new bone formation in AS animal models, while overexpression of CXCL12 led to joint ankylosis phenotype. Rac1 was found essential for OPCs migration and pathological new bone formation. These findings suggest a potential strategy for targeting the CXCL12/ CXCR4-Rac1 axis to prevent progression of axial skeleton ankylosis in AS.

IL-17A expressed on neutrophil extracellular traps promotes mesenchymal stem cell differentiation toward bone-forming cells in ankylosing spondylitis

Charalampos Papagoras et al.

Summary: The research found that neutrophil extracellular traps (NETs) formation was enhanced in patients with ankylosing spondylitis. NETs decorated with interleukin-17A (IL-17A) stimulated the differentiation of bone marrow mesenchymal stem cells (MSCs) towards bone-forming cells. The expression of IL-17A was regulated by IL-1 beta, and blocking the IL-1 beta signaling pathway disrupted osteogenesis driven by IL-17A-bearing NETs.

microRNA-214-3p Suppresses Ankylosing Spondylitis Fibroblast Osteogenesis via BMP-TGFβ Axis and BMP2

Lixiang Ding et al.

Summary: MicroRNA miR-214-3p plays a significant role in fibroblast osteogenesis in ankylosing spondylitis (AS) by targeting BMP2 and blocking the BMP-TGF beta axis, providing a potential novel insight for AS treatment.
