Metabolism of tissue macrophages in homeostasis and pathology

Stefanie K. Wculek et al.

Summary: Cellular metabolism plays a crucial role in maintaining and regulating immune cell activities, with macrophages being essential for tissue homeostasis and potentially contributing to pathological conditions. Different macrophage populations in various organs have distinct functions, posing metabolic challenges for them to fulfill their roles in specific microenvironments.

Phagosome-regulated mTOR signalling during sarcoidosis granuloma biogenesis

Elliott D. Crouser et al.

Summary: Comparing the molecular characteristics of granulomas formed in response to Mycobacterium tuberculosis antigens in sarcoidosis patients and those with latent tuberculosis infection (LTBI) revealed differences in genomic expression. The assembly and acidification of phagolysosomes were found to support mTORc1 signaling and promote granuloma formation in sarcoidosis.

Cancer-associated fibroblasts and the related Runt-related transcription factor 2 (RUNX2) promote bladder cancer progression

Bitian Liu et al.

Summary: Research indicates that high infiltration of CAFs is associated with tumor progression and poor prognosis in BLCA. RUNX2, an overexpressed transcription factor in bladder cancer, is related to CAFs and affects prognosis. Therefore, RUNX2 may serve as a potential marker related to CAFs and a therapeutic target in BLCA.

Treatment of granuloma annulare and suppression of proinflammatory cytokine activity with tofacitinib

Alice Wang et al.

Summary: Single-cell RNA sequencing revealed upregulation of IFN-gamma production by CD41 T cells in GA lesions, which is associated with inflammatory polarization of macrophages and fibroblasts. Treatment with tofacitinib inhibited IFN-gamma, oncostatin M, IL-15, and IL-21 activity, leading to clinical and histologic disease remission in some patients and marked improvement in others. The Janus kinase-signal transducer and activator of transcription pathway is activated in GA, and Janus kinase inhibitors appear to be an effective treatment option.

Summary: As the number of single-cell transcriptomics datasets increases, integrating data to achieve a common ontology of cell types and states becomes the natural next step. scVI and scANVI are two methods that can effectively integrate data and perform well in cell state annotation, with high accuracy, scalability, and adaptability.

Developmental cell programs are co-opted in inflammatory skin disease

Gary Reynolds et al.

Summary: The skin establishes a complex cellular network early in embryonic development to provide biophysical and immunological protection. The study found an enrichment of innate immune cells in skin during the first trimester, as well as clonal expansion of disease-associated lymphocytes in atopic dermatitis and psoriasis. Additionally, a reemergence of prenatal vascular endothelial cell and macrophage cellular programs was observed in atopic dermatitis and psoriasis lesional skin.

Fibroblasts: Origins, definitions, and functions in health and disease

Maksim Plikus et al.

Summary: Fibroblasts are diverse mesenchymal cells that play a role in tissue homeostasis and disease, producing complex extracellular matrix and creating signaling niches. They are transcriptionally and functionally heterogeneous across and within organs, encoding positional information and maintaining distinct cellular progeny. Their unique properties make them poised for tissue repair but can also drive fibrotic disorders if activated aberrantly.

Transcriptomics of bronchoalveolar lavage cells identifies new molecular endotypes of sarcoidosis

Milica Vukmirovic et al.

Summary: The study identified specific gene expression patterns in bronchoalveolar lavage (BAL) cells that characterize major pulmonary phenotypes of sarcoidosis, suggesting the presence of distinct disease molecular endotypes. Major associations were found between transcriptional programs and pulmonary manifestations of sarcoidosis, as well as potential endotypes reflecting different patterns of organ involvement. Validation of these endotypes was performed using gene expression data, pulmonary function tests, and cell differentials from an independent sarcoidosis cohort.

CD4+ T cells in the lungs of acute sarcoidosis patients recognize an Aspergillus nidulans epitope

Sarah A. Greaves et al.

Summary: The acute form of sarcoidosis known as Lofgren's syndrome is genetically associated with HLA-DR3 and characterized by an accumulation of CD4(+) T cells in bronchoalveolar lavage. Research has identified a peptide from Aspergillus nidulans that stimulates CD4(+) T cells in an HLA-DR3-restricted manner in LS patients. Additionally, LS subjects showed increased IFN-gamma and IL-2-secreting T cells in response to the NDPD peptide, along with elevated IgG antibody responses to A. nidulans NDPD in their serum. These findings suggest a potential role of A. nidulans in the etiology of LS.

The cellular architecture of the antimicrobial response network in human leprosy granulomas

Feiyang Ma et al.

Summary: The study identified a set of genes involved in antimicrobial responses that are differentially expressed in reversal reactions (RRs) lesions compared to lepromatous leprosy (L-lep) lesions, regulated by interferon-gamma and interleukin-1 beta. By integrating spatial coordinates of key cell types and antimicrobial gene expression, a map depicting the organized architecture of granulomas in RR and T-lep lesions was constructed, showing compositional and functional layers contributing to the antimicrobial response. Modlin and colleagues utilized single-cell RNA sequencing with cellular spatial mapping to examine the architecture of granulomas in leprosy lesions, providing insights from localized disease (tuberculoid leprosy, reversal reaction) to progressive infection (lepromatous leprosy).

Inhibition of Granuloma Triglyceride Synthesis Imparts Control of Mycobacterium tuberculosis Through Curtailed Inflammatory Responses

Stanzin Dawa et al.

Summary: Lipid metabolism plays a crucial role in the interaction between host and pathogen during Mycobacterium tuberculosis infection. The accumulation of triglyceride in necrotic granulomas is important for the success of the pathogen, and manipulating lipid droplet homeostasis may potentially offer a new approach for host-directed therapy in Tuberculosis.
