SIRT1 regulates trophoblast senescence in premature placental aging in preeclampsia

YuJia Wang et al.

Summary: The downregulation of SIRT1 and the upregulation of senescence- and ECM-related proteins are observed in premature aging placenta with preeclampsia. Activation of SIRT1 can abolish senescence in syncytialized BeWo cells, while knockdown of SIRT1 restores these effects.

Computational analysis identified accelerated senescence as a significant contribution to preeclampsia pathophysiology

Nadia Siddique et al.

Summary: This study demonstrates the significant role of placental senescence in the pathogenesis of preeclampsia. The mechanism of senescence varies among different subtypes of preeclampsia, which could serve as a basis for identifying potential drug targets.

SRT1720 plays a role in oxidative stress and the senescence of human trophoblast HTR8/SVneo cells induced by D-galactose through the SIRT1/ FOXO3a/ROS signalling pathway

Lanlan Yin et al.

Summary: This study evaluated the effects of D-gal exposure on HTR8/SVneo cells and its underlying mechanism. D-gal was found to decrease the activity of HTR8/SVneo human trophoblasts and induce premature senescence, which was mediated by oxidative stress. The SIRT1 activator SRT1720 mitigated D-gal-induced oxidative stress and senescence by upregulating SIRT1 and FOXO3a expression and reducing ROS production.

SIRT1: A Novel Protective Molecule in Pre-eclampsia

Zhenzhen Liu et al.

Summary: Pre-eclampsia is a severe complication of pregnancy characterized by insufficient trophoblast invasion, impaired uterine spiral artery remodeling, placental hypoxia and ischemia, and endothelial dysfunction. The role of SIRT1, a NAD+-dependent deacetylase, in pre-eclampsia and its mechanisms have been explored in recent studies. SIRT1 seems to affect placental development and trophoblast invasion, while protecting vascular endothelial cells from oxidative stress, inflammatory response, autophagy, and senescence. Activation of SIRT1 using compounds such as resveratrol has shown potential in attenuating symptoms of pre-eclampsia.

Inhibition of miR-494-3p alleviates oxidative stress-induced cell senescence and inflammation in the primary epithelial cells of COPD patients

Qinghua Zeng et al.

Summary: MiR-494-3p expression is upregulated by oxidative stress through the p38MAPK-c-myc signaling pathway, leading to direct binding to SIRT3 and reduction of its expression, resulting in increased cellular senescence and contributing to COPD progression.

Summary: The study found that markers of placental oxidative stress and senescence increase in post-term pregnancies and cases of pre-eclampsia. As gestational age increases, antioxidant defenses decrease, providing insight into placental health and potentially aiding in determining the optimal gestational age for delivery.
