A conserved role of the Hippo signalling pathway in initiation of the first lineage specification event across mammals

Claudia Gerri et al.

Summary: Our study investigates the molecular events driving cell specification in early mammalian development, specifically focusing on the role of aPKC and Hippo signaling. We find that the establishment of cell polarity via aPKC is conserved in mouse, cow, and human embryos. Inhibition of the Hippo pathway leads to ectopic TE initiation and downregulation of SOX2 in all four species. However, there are differences in timing and localization of molecular markers, with rat embryos showing closer resemblance to human and cow embryos.

Entropy sorting of single-cell RNA sequencing data reveals the inner cell mass in the human pre-implantation embryo

Arthur Radley et al.

Summary: A mathematical framework called entropy sorting (ES) is introduced to address the challenge of discerning meaningful cellular heterogeneity from noise in single-cell gene expression analysis. ES quantifies observed correlations between features to distinguish genes indicative of cell identity in an unsupervised manner, without the need for user-defined significance thresholds.

Human embryo polarization requires PLC signaling to mediate trophectoderm specification

Meng Zhu et al.

Summary: The apico-basal polarization of cells in the human embryo occurs in two steps, involving the enrichment of F-actin and the apical accumulation of the PAR complex. The formation of an apical domain relies on PLC signaling, specifically the enzymes PLCB1 and PLCE1, from the eight-cell stage. This polarization process is crucial for reinforcing a trophectoderm fate and regulating the first lineage segregation in human embryos.

Integrated pseudotime analysis of human pre-implantation embryo single-cell transcriptomes reveals the dynamics of lineage specification

Dimitri Meistermann et al.

Summary: By analyzing single-cell RNA sequencing data and time-lapse imaging, we investigated the transcriptomic changes during early human embryo development. Our study identified divergence in trophectoderm/inner cell mass transcriptomes between the B2 to B3 blastocyst stages and highlighted potential cell fate markers.

Modelling human blastocysts by reprogramming fibroblasts into iBlastoids

Xiaodong Liu et al.

Summary: The study describes a method to reprogram fibroblasts into human blastocyst models, known as iBlastoids. This model can mimic the overall structure of blastocysts, including inner cell mass, epiblast, primitive endoderm-like cells, blastocoel-like cavity, and trophectoderm-like cells. The iBlastoids system is believed to facilitate the study of early human development, the effects of gene mutations and toxins during early embryogenesis, and the development of new therapies associated with in vitro fertilization.

Blastocyst-like structures generated from human pluripotent stem cells

Summary: The study developed an effective three-dimensional culture strategy to generate blastocyst-like structures in vitro which resemble human blastocysts. These structures, termed 'human blastoids', are readily accessible, scalable, versatile, and perturbable alternative to blastocysts for studying early human development, understanding early pregnancy loss, and gaining insights into early developmental defects.

Embryo selection with artificial intelligence: how to evaluate and compare methods?

Mikkel Fly Kragh et al.

Summary: Embryo selection in IVF involves evaluating fertilized egg quality and using AI to assist in selecting embryos most likely to result in successful pregnancies. Debate exists on whether AI methods outperform manual evaluation, with variations in evaluation metrics and comparisons across studies.
