Functional characterization of two variants of mitochondrial topoisomerase TOP1MT that impact regulation of the mitochondrial genome

Iman Al Khatib et al.

Summary: This study identified two variants of TOP1MT that are associated with hypertrophic cardiomyopathy and impair the function of TOP1MT. These findings provide insights into the roles of TOP1MT in maintaining and expressing the mitochondrial genome and suggest that impairments in this protein may contribute to human pathology.

DNA sensing and associated type 1 interferon signaling contributes to progression of radiation-induced liver injury

Shisuo Du et al.

Summary: Liver damage caused by exposure to ionizing radiation can lead to liver dysfunction, particularly in cases of radiotherapy for cancers such as hepatocellular carcinoma. Experimental studies have shown that activation of the cGAS-STING pathway in non-parenchymal cells plays a key role in the development of radiation-induced liver disease (RILD) by triggering interferon production and release. Targeting the interferon signaling pathway may offer a potential protective strategy against RILD.

Loss of Mitochondrial Protease CLPP Activates Type I IFN Responses through the Mitochondrial DNA-cGAS-STING Signaling Axis

Sylvia Torres-Odio et al.

Summary: CLPP deficiency results in abnormal immune system responses in mice, leading to increased resistance to RNA and DNA virus infections. The instability and packaging alterations of mitochondrial DNA caused by CLPP deficiency drive the IFN-I signaling pathway.

cGAS-STING cytosolic DNA sensing pathway is suppressed by JAK2-STAT3 in tumor cells

Manuel Adrian Suter et al.

Summary: Deficiencies in DNA repair and nucleases lead to cytosolic DNA accumulation. The cGAS-STING pathway is important for detecting cytosolic DNA, but its function can be modulated by IL-6 and JAK2/STAT3 signaling pathways in cancer cells, affecting interferon expression and tumor rejection.

STING Operation at the ER/Golgi Interface

Tomohiko Taguchi et al.

Summary: DNA is present in the nucleus and mitochondria of eukaryotic cells, but can also be found in the cytosol in certain instances. The cytosolic DNA consists of self DNA leaked from the nucleus or mitochondria, as well as non-self DNA from DNA viruses, which triggers the host immune response. Recent studies have identified key molecules, cGAS and STING, in this immune response, with STING playing a crucial role in triggering various cellular responses.

Mitochondria-Induced Immune Response as a Trigger for Neurodegeneration: A Pathogen from Within

Marta Luna-Sanchez et al.

Summary: The symbiosis between mitochondria and the ancestor of the eukaryotic cell is crucial for cellular complexity and survival, but compromised mitochondrial integrity can lead to unwanted cellular immune responses. Mitochondrial nucleic acids like mtDNA and mtRNA can interact with cytoplasmic sensors, triggering inflammation. Mitochondria-driven inflammation may serve as a potential therapeutic target for neurodegenerative and primary mitochondrial diseases.

Enhanced cGAS-STING-dependent interferon signaling associated with mutations in ATAD3A

Alice Lepelley et al.

Summary: Mutations in the mitochondrial membrane protein ATAD3A lead to enhanced interferon signaling, resulting in a novel type I interferonopathy. Knockdown of ATAD3A can suppress interferon signaling. This sheds light on the role of mitochondrial membrane protein ATAD3A in immune system activation.

Cytosolic dsDNA of mitochondrial origin induces cytotoxicity and neurodegeneration in cellular and zebrafish models of Parkinson's disease

Hideaki Matsui et al.

Summary: This study suggests that mitochondrial dysfunction and lysosomal dysfunction play a role in the pathogenesis of Parkinson's disease. The leakage of mitochondrial DNA into the cytosol induces cytotoxicity and neurodegeneration in cellular and zebrafish models of PD. Knock-down of IFI16, a cytosolic dsDNA sensor, or overexpression of lysosomal DNase II can rescue this cytotoxicity.

Summary: In the mutator mouse model with mitochondrial DNA instability, aberrant activation of the IFN-I immune axis leads to immunometabolic dysfunction, shortened lifespan, and accelerated aging. Elevated IFN-I signaling suppresses activation of NRF2, increasing oxidative stress and proinflammatory responses, resulting in metabolic dysfunction.

Mitochondrial Nucleic Acid as a Driver of Pathogenic Type I Interferon Induction in Mendelian Disease

Alice Lepelley et al.

Summary: The immune response to viral infection involves the detection of pathogen-derived nucleic acids, but it is crucial to distinguish self from non-self nucleic acids. Mitochondria are recognized as a potential source of immunogenic self nucleic acids, and their release into the cytosol can lead to abnormal IFN induction.
