Metabolic reprogramming of the intestinal microbiome with functional bile acid changes underlie the development of NAFLD

Ekaterina Smirnova et al.

Summary: This study demonstrates the significance of bile acids and the bile acid dependent microbiome in the development and progression of NAFLD, and suggests their potential use as biomarkers and therapeutic targets for NASH.

A multiancestry genome-wide association study of unexplained chronic ALT elevation as a proxy for nonalcoholic fatty liver disease with histological and radiological validation

Marijana Vujkovic et al.

Summary: Through a multi-ancestry genome-wide association study, we identified multiple genetic loci associated with NAFLD in the Million Veteran Program, some of which are also related to metabolic and/or inflammatory traits. Integrating cALT, histology, and imaging provided new insights into genetic susceptibility to NAFLD.

Low real-world eligibility rates for nonalcoholic steatohepatitis patients for phase 3 clinical trial criteria application in a large academic health system

Ysabel C. Ilagan-Ying et al.

Summary: In a university-based cohort of NASH patients, few met the enrollment criteria for phase 3 clinical trials, mostly due to low rates of liver biopsy. Among those with histologic confirmation, 14.4% met the enrollment criteria. Validation of generalizability for safety and efficacy of investigational NASH agents in real-world populations is necessary.

Prospective Study of Outcomes in Adults with Nonalcoholic Fatty Liver Disease

Arun J. Sanyal et al.

Summary: In this study involving patients with nonalcoholic fatty liver disease, advanced fibrosis stages F3 and F4 were associated with increased risks of liver-related complications and death over a median of 4 years of follow-up. Advanced fibrosis was also related to an increased incidence of liver decompensation events.

Bile acids profile, histopathological indices and genetic variants for non-alcoholic fatty liver disease progression

Nisreen Nimer et al.

Summary: Metabolomic studies have shown that plasma levels of bile acids are elevated in subjects with NAFLD compared to healthy controls. Specific bile acids are associated with the transition from simple hepatic steatosis to NASH, as well as genetic determinants. This suggests a potential role for bile acid metabolism in the development and progression of NASH.

Mechanisms and disease consequences of nonalcoholic fatty liver disease

Rohit Loomba et al.

Summary: NAFLD and NASH are leading chronic liver diseases worldwide, causing progressive liver injury that can lead to cirrhosis and hepatocellular carcinoma. These diseases are associated with hepatic glucose and lipid metabolism, and are influenced by genetic, epigenetic, and environmental factors.

Bile Acids, Liver Cirrhosis, and Extrahepatic Vascular Dysfunction

Tilman Sauerbruch et al.

Summary: This review examines the role of bile acids in the hyperdynamic circulatory disturbance of liver cirrhosis, and suggests potential ways to influence hemodynamic disorder through modulation of the bile acid pool or dysbiosis. Limited evidence and lack of long-term studies exist on this topic.
