Antibody signatures in inflammatory bowel disease: current developments and future applications

Arno R. Bourgonje et al.

Summary: The adequate functioning of the intestinal barrier is crucial for protecting against pathogens and tolerating microbiota and self-antigens. Inflammatory bowel diseases (IBDs) are characterized by a disrupted intestinal barrier integrity, resulting in an abnormal immune response. Recent innovative antibody profiling technologies have the potential to provide a comprehensive understanding of humoral immunity and its applications in disease diagnosis, therapy, and prevention in IBD.

Novel Fecal Biomarkers That Precede Clinical Diagnosis of Ulcerative Colitis

Heather J. Galipeau et al.

Summary: Increased fecal proteolytic activity was found before the clinical diagnosis of UC, which may serve as a noninvasive biomarker of inflammation associated with changes in gut microbiota. The study suggests potential therapeutic targets for at-risk populations based on antiprotease treatments.

Serum proteomic profiling at diagnosis predicts clinical course, and need for intensification of treatment in inflammatory bowel disease

R. Kalla et al.

Summary: Personalized medicine success in complex diseases relies on biomarker discovery. A study identified serum proteins distinguishing IBD from non-IBD, as well as predicting escalation with specific protein combinations. The research also highlighted proteins associated with genetic variations, urging further validation in prospective studies.

Summary: Prostate cancer is a leading cause of morbidity and mortality among men worldwide, and advancements in multiple protein biomarkers offer hope for its diagnosis and prognosis. The identification of multi-protein biomarkers with high sensitivity and specificity, as well as their integration with clinicopathologic parameters, imaging, and other molecular biomarkers, can help achieve optimal personalized management for PC patients.

Oncostatin M Is a Biomarker of Diagnosis, Worse Disease Prognosis, and Therapeutic Nonresponse in Inflammatory Bowel Disease

Sare Verstockt et al.

Summary: The study found that Oncostatin M (OSM) can serve as a diagnostic biomarker in the tissue and serum of IBD patients, as well as predict prognosis and response to biologic therapies. Elevated ileal OSM/OSMR levels were significantly associated with postoperative recurrent CD. Serum OSM levels were increased in newly diagnosed patients and postoperative patients with recurrent CD. In families with IBD, higher serum levels were observed in first-degree relatives (FDRs) than in control families.

TREM-1+ Macrophages Define a Pathogenic Cell Subset in the Intestine of Crohn's Disease Patients

Charles Caer et al.

Summary: The study showed that high expression of intestinal TREM-1 in IBD patients is associated with a high frequency of TREM-1(+) immature macrophages and TREM-1(+)CD11b(+) granulocytes, which contribute to the deleterious inflammatory microenvironment. Blocking the TREM-1 pathway, especially simultaneously with anti-TNF therapy, has potential as a new therapy for IBD.

Matrix Metalloproteinase MMP-12 Promotes Macrophage Transmigration Across Intestinal Epithelial Tight Junctions and Increases Severity of Experimental Colitis

Meghali Nighot et al.

Summary: This study demonstrates the important role of MMP-12 in intestinal TJ barrier function and inflammation, showing that MMP-12(-/-) mice have increased tolerance to DSS colitis. Macrophage transmigration across the epithelial layer is MMP-12 dependent, highlighting MMP-12-mediated degradation of basement membrane laminin as a key pathogenic factor for intestinal inflammation.

Serum biomarkers confirming stable remission in inflammatory bowel disease

Christoph Kessel et al.

Summary: Crohn's disease (CD) and ulcerative colitis (UC) have a chronic-remittent course. Optimal management of inflammatory bowel diseases (IBD) relies on early intervention, treat-to-target strategies and a tight disease control. However, it is challenging to assess the risk of relapses in individual patients. We investigated blood-based biomarkers for the confirmation of disease remission in patients with IBD. Our study identified serum biomarkers with the potential to indicate unstable disease control in IBD, which may help to steer individual therapies to maintain remission.

Anti-Microbial Antibody Response is Associated With Future Onset of Crohn's Disease Independent of Biomarkers of Altered Gut Barrier Function, Subclinical Inflammation, and Genetic Risk

Sun-Ho Lee et al.

Summary: High baseline total positive antibody response is associated with an increased risk of developing CD in the future, independently of abnormal gut barrier function, subclinical inflammation, and CD-related genetic risks. Causal mediation analysis showed that the effect of high AS on future CD development is partially mediated through preclinical gut inflammation.

Systemic Inflammation in Preclinical Ulcerative Colitis

Daniel Bergemalm et al.

Summary: In preclinical ulcerative colitis, several inflammatory proteins are up-regulated years before diagnosis, which can effectively predict the risk of developing ulcerative colitis. These proteins are highly indicative of an ulcerative colitis diagnosis and show early upregulation upon exposure to genetic and environmental risk factors.
