Molecular Mechanisms and Regulation of Mammalian Mitophagy

Vinay Choubey et al.

Summary: Mitochondria play a crucial role in energy production, biomolecule synthesis, and cell fate determination. To overcome challenges like oxidative stress, protein-related issues, and mitochondrial DNA damage, mitochondria have robust quality control mechanisms and utilize mitophagy to clear damaged mitochondria. This review provides an in-depth overview of mammalian mitophagy, its importance in human health and diseases, and future research directions in this field.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

WNT/β-catenin-suppressed FTO expression increases m6A of c-Myc mRNA to promote tumor cell glycolysis and tumorigenesis

Xueying Yang et al.

Summary: FTO plays a critical role in cancer development by removing m(6)A modifications from genes. Downregulation of FTO expression in lung adenocarcinoma patients is associated with poor survival. Mechanistically, Wnt signaling induces EZH2 binding to beta-catenin, inhibiting FTO expression and promoting m(6)A modifications on specific genes like MYC to enhance tumor cell glycolysis and growth.
