DNA hypermethylation modification promotes the development of hepatocellular carcinoma by depressing the tumor suppressor gene ZNF334

Dapeng Sun et al.

Summary: DNA methylation plays a crucial role in the development and progression of hepatocellular carcinoma (HCC). This study systematically illustrated the dynamic changes in DNA methylation from premalignant to early-stage HCC. The findings showed that DNA hypermethylation of tumor suppressor genes was predominant in early-stage HCC, particularly affecting the expression of ZNF334. Moreover, high expression of ZNF334 was associated with longer overall survival and relapse-free survival in patients. Overall, this study provides important insights into the role of DNA methylation in HCC and suggests ZNF334 as a potential target for early diagnosis and precise treatment.

Attenuated expression of SNF5 facilitates progression of bladder cancer via STAT3 activation

Hua Ding et al.

Summary: The study revealed that low SNF5 expression in bladder cancer is associated with poor prognosis and N-stage, promoting cell proliferation and migration through STAT3 activation. Additionally, low SNF5 expression renders cells sensitive to EGFR inhibitor gefitinib and combination therapy of GSK126 and cisplatin.

Cancer-related SRCAP and TPR mutations in colon cancers

Seong Won Moon et al.

Summary: The study identified frameshift mutations of SRCAP, TPR, and CEACAM5 genes in high microsatellite instability colon cancers, with regional differences in the mutations and increased SRCAP expression. These findings could contribute to the development of colon cancer with high microsatellite instability.

BRG1 knockdown inhibits proliferation through multiple cellular pathways in prostate cancer

Katherine A. Giles et al.

Summary: The study found that BRG1 is overexpressed in prostate cancer and mainly affects global transcription programs by down-regulating gene expression, with most downregulated genes related to proliferation. Depletion of BRG1 results in G1 arrest.

PRMT1-mediated EZH2 methylation promotes breast cancer cell proliferation and tumorigenesis

Zhongwei Li et al.

Summary: This study uncovers the critical role of PRMT1-mediated methylation of EZH2 at the R342 site in promoting breast cancer cell proliferation, accelerating cell cycle progression, repressing P16 and P21 transcription, and enhancing EZH2 binding with SUZ12 and PRC2 assembly. Additionally, it highlights the potential of meR342-EZH2 as a novel therapeutic target for breast cancer treatment, indicating a negative correlation with pT311-EZH2 expression.

LSD1 Promotes Prostate Cancer Cell Survival by Destabilizing FBXW7 at Post-Translational Level

Xu-ke Qin et al.

Summary: Prostate cancer is the most common cancer in men worldwide, and castration-resistant prostate cancer is incurable and prone to drug resistance. Up-regulated LSD1 in prostate cancer can destabilize cancer suppressor protein FBXW7 without its demethylation function, promoting cancer cell survival. Inhibition of LSD1-FBXW7 interaction by the allosteric inhibitor SP-2509 can suppress cancer cell survival, suggesting a potential new direction for prostate cancer treatment research.

The impact of CBP expression in estrogen receptor-positive breast cancer

Wafaa S. Ramadan et al.

Summary: The study found that elevated expression of CBP and GCN5 in breast cancer patients and cell lines, especially in luminal A and B subtypes. Analysis of publicly available databases showed amplification of CREBBP and KAT2A genes in breast cancer patients. Patients with high CREBBP or KAT2A gene expression had better 5-year disease-free survival than the low gene expression group.

AID in Chronic Lymphocytic Leukemia: Induction and Action During Disease Progression

Pablo Oppezzo et al.

Summary: This review article discusses the dual role of the enzyme activation-induced cytidine deaminase (AID) in adaptive immunity and B-cell malignancies, emphasizing its pathological effects as a causative factor in leukemia and lymphomas. It summarizes the expression and function of AID in normal B lymphocytes, evaluates potential causes for AID expression in leukemic cells, and discusses its role in lymphomagenesis based on recent genomic landscape analyses of leukemia and lymphomas. The review also touches upon the correlation between AID off-target mutations and tumor-gene drivers in these cancers, and how these mutations could impact disease progression.

Summary: The study revealed that in patients with oral tongue squamous cell carcinoma, overexpression of UTX was significantly associated with poorer survival outcomes, suggesting that UTX plays a critical role in the progression of OTSCC. The inhibition of UTX with GSK-J4 also showed promising results in suppressing cancer migration and invasion.

Sherry X. Yang et al.

Summary: This study examined the effects of T-dCyd treatment on cancer cell lines with TET2 and DNMT3A mutations, finding significant antitumor activity in cells with these mutations compared to those without. Furthermore, T-dCyd treatment led to inhibition of tumor growth in xenograft models with concurrent TET2 and DNMT3A mutations, suggesting sensitivity to the drug in these cases. The study also identified co-mutations of TET2 and DNMT3A in human solid tumors and specific hematological malignancies.

Misregulation of the expression and activity of DNA methyltransferases in cancer

Isaiah K. Mensah et al.

Summary: DNA methyltransferases play a crucial role in maintaining DNA methylation in mammalian cells, with mutations and abnormal expression affecting cell proliferation and identity in cancer. Aberrant DNA methylation is a hallmark of most cancers, and despite the effectiveness of DNMT inhibitors in some cases, their lack of specificity can lead to a range of deleterious effects. Understanding the distinct molecular mechanisms of DNMTs could help in identifying alternative cancer therapeutic targets.
