Bioinformatics analysis based on high-throughput sequencing data to identify hub genes related to different clinical types of COVID-19

Shengying Liu et al.

Summary: This study aimed to explore hub genes related to different clinical types of COVID-19 cases and predict therapeutic drugs for severe cases. The expression profile of GSE166424 was divided into four data sets based on clinical types of COVID-19, and differential expression genes (DEGs) were calculated. Specific genes for each clinical type were identified and subjected to enrichment analysis, protein-protein interaction (PPI) network analysis, hub gene screening, and ROC curve analysis. Hub genes related to severe cases were verified in GSE171110, and their tissue specificity was obtained from the HPA database. Potential therapeutic drugs were predicted using the DGIdb database. The study identified specific genes for asymptomatic infections, mild, moderate, and severe cases, and discovered potential diagnostic biomarkers for severe COVID-19. Additionally, 55 drugs were found to have potential therapeutic value for severe COVID-19.

Hypoxia Increases the Potential for Neutrophil-mediated Endothelial Damage in Chronic Obstructive Pulmonary Disease

Katharine M. Lodge et al.

Summary: Patients with chronic obstructive pulmonary disease (COPD) have a high incidence and mortality rate of cardiovascular diseases, and exacerbations further increase the risk. Hypoxia drives the release of neutrophil elastase, leading to tissue injury. This study found that under hypoxic conditions, COPD neutrophils release more elastase and cause damage to pulmonary endothelial cells, as well as increased adherence to endothelial cells. Plasma from COPD patients also showed higher levels of hypoxia-upregulated neutrophil-derived proteins and protease activity, indicating vascular injury.

A multi-tissue study of immune gene expression profiling highlights the key role of the nasal epithelium in COVID-19 severity

Alberto Gomez-Carballa et al.

Summary: This study analyzed the gene expression of immune-related genes in different tissues (nasal, buccal, and blood) from patients with different severities of COVID-19 and healthy controls. The findings suggest that mild and asymptomatic cases exhibit a powerful innate antiviral response in the nasal epithelium, while severe cases lack this response. Severe cases show signs of viral activity and viral dissemination to the lower respiratory tract in the oral mucosa, which may explain the exacerbated innate immune response and impaired adaptive immune response observed at a systemic level. The results also suggest the potential role of the buccal cavity in SARS-CoV-2 infection and dissemination in patients with worse prognosis.

Tet2 deficiency drives liver microbiome dysbiosis triggering Tc1 cell autoimmune hepatitis

Surya P. Pandey et al.

Summary: This study reveals that lack of hematopoietic Tet methylcytosine dioxygenase 2 (Tet2) contributes to the development of autoimmune hepatitis (AIH). AIH is associated with interferon-gamma and aryl hydrocarbon receptor (AhR) signaling pathways, as well as the presence of AhR ligand-producing pathobionts in the liver. Furthermore, the study demonstrates that the AhR ligand indole-3aldehyde (I3A) released by Lactobacillus reuteri can trigger AIH-like pathology.

Human seasonal influenza under COVID-19 and the potential consequences of influenza lineage elimination

Vijaykrishna Dhanasekaran et al.

Summary: Since the onset of the COVID-19 pandemic, public health measures and travel restrictions have heavily suppressed global influenza circulation. The genetic diversity of seasonal influenza viruses has significantly decreased, with the influenza B/Yamagata lineage not conclusively detected. Travel restrictions have confined the circulation of different influenza viruses to specific regions, leading to a continuous decline in global transmission of seasonal influenza, except in select hotspots that may seed future epidemics. Additionally, the selection of influenza vaccine strains and epidemic control face challenges due to waning population immunity and sporadic case detection.

IL-36 receptor agonist and antagonist imbalance drives neutrophilic inflammation in COPD

Jonathan R. Baker et al.

Summary: Current treatments for chronic obstructive pulmonary disease (COPD) are unable to modify the underlying pathophysiology and disease progression, necessitating the search for alternative therapies. This study reveals that COPD patients have increased levels of IL-36α and decreased levels of IL-36 receptor antagonist (IL-36Ra) in bronchoalveolar and nasal fluid. IL-36α stimulates the release of CXCL1, which can be inhibited by exogenous IL-36Ra, suggesting that blocking IL-36R could attenuate COPD-induced inflammation.

Influenza A induces lactate formation to inhibit type I IFN in primary human airway epithelium
