Oriol Pich et al.

Summary: The translational challenges in the field of precision oncology are partly due to the complexity and diversity of cancer. Advances in genomics have enabled large-scale sequencing efforts and discoveries, impacting our understanding of cancer initiation, prevention, treatment, and other concepts. Key areas for improving cancer outcomes are discussed, along with the practical challenges of adopting molecular profiling in clinical settings and the need for robust translational infrastructure.

Amino acid transporter LAT1 (SLC7A5) as a molecular target for cancer diagnosis and therapeutics

Yoshikatsu Kanai

Summary: Cancer cells require a massive supply of nutrients, including amino acids. LAT1 is an amino acid transporter that is specifically expressed in cancer cells. High expression of LAT1 is associated with poor prognosis in patients. Inhibition of LAT1 can suppress cancer cell proliferation and tumor growth. LAT1 can also be used as a target for cancer diagnosis and specific delivery of anti-tumor drugs.

Single-nucleus cross-tissue molecular reference maps toward understanding disease gene function

Gokcen Eraslan et al.

Summary: Understanding the function and regulation of genes in homeostasis and disease requires knowledge of their expression in different cellular and tissue contexts. In this study, we performed single-nucleus RNA sequencing on various frozen tissue types, generating a cross-tissue atlas of cellular profiles. By integrating this atlas, we identified shared and tissue-specific features of cell populations, as well as cell types and gene modules associated with neuromuscular, metabolic, and immune components of monogenic diseases and complex traits analyzed by genome-wide association studies.

A transmembrane protein family gene signature for overall survival prediction in osteosarcoma

Yuehui Du et al.

Summary: We constructed a prognostic signature based on six TMEM protein family genes and found that it can stratify osteosarcoma patients into high- and low-risk groups. Functional enrichment analysis revealed the association between immunity and the prognostic signature. Additionally, the high-risk group had lower proportions of M2 macrophages in the immune microenvironment and lower scores of neutrophils and Treg.

Cell Division Cycle-Associated Protein 3 (CDCA3) Is a Potential Biomarker for Clinical Prognosis and Immunotherapy in Pan-Cancer

Yingkun Xu et al.

Summary: CDCA3 is an essential regulator in cell mitosis and its dysregulation is common in human cancers. Our comprehensive analysis using multiple cancer-related databases and bioinformatics tools revealed that CDCA3 is overexpressed in almost all cancer types and is significantly associated with survival in multiple cancer types. CDCA3 expression positively correlates with immune cell infiltration levels in human cancers. Moreover, overexpression of CDCA3 may promote the malignant progression of cancer by activating various oncogenic signaling pathways.

Summary: Efforts to develop anti-cancer therapies have mainly targeted the epithelial compartment, but recent studies have shown the significant influence of cancer-associated fibroblasts (CAFs) in tumor progression. CAFs not only promote cancer cell proliferation, therapy resistance, and immune exclusion, but may also restrain tumor progression in certain contexts. Research on CAFs has focused on their heterogeneity, plasticity, and functions across different cancer types and states, as well as advancements in therapeutic strategies targeting CAFs currently undergoing preclinical and clinical evaluation.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
