Transcription factor SOX15 regulates stem cell pluripotency and promotes neural fate during differentiation by activating the neurogenic gene Hes5

Summary: SOX2 and SOX15 are Sox family transcription factors that play important roles in embryonic stem cells (ESCs). While the role of SOX2 in stem cell self-renewal and pluripotency acquisition is well-known, the contribution of SOX15 is still unclear. However, studies have shown that overexpression of SOX15 can partially compensate for the self-renewal defects in SOX2-deficient ESCs. This study reveals that SOX15 contributes to stem cell maintenance by working with coactivators to ensure optimal expression of pluripotency-related genes. SOX15 also plays a role in neural lineage specification, as its loss leads to defects in the commitment of ESCs to neural fates.
