The interplay between BAX and BAK tunes apoptotic pore growth to control mitochondrial-DNA-mediated inflammation

Katia Cosentino et al.

Summary: BAX and BAK are important regulators of apoptosis and play a role in the formation of apoptotic pores. BAK has faster kinetics in forming smaller structures and can accelerate the assembly of BAX. BAX and BAK regulate each other during the formation of apoptotic pores, which affects the release kinetics of mitochondrial components and the activation kinetics of the cGAS/STING pathway.

Maltol Promotes Mitophagy and Inhibits Oxidative Stress via the Nrf2/PINK1/Parkin Pathway after Spinal Cord Injury

Yuqin Mao et al.

Summary: The study found that maltol can trigger the expression of Nrf2 and promote its retranslocation to the nucleus, thereby obstructing OS signal and apoptosis-mediated neuronal cell death after SCI. Additionally, maltol treatment enhances PINK1/Parkin-mediated mitophagy in PC12 cells, aiding in the recovery of mitochondrial functions. These findings suggest that maltol could be a promising therapeutic candidate for the treatment and management of SCI.

Activation of PINK1/Parkin-mediated mitophagy protects against apoptosis in kidney damage caused by aluminum

Pengli Liu et al.

Summary: The study found that aluminum induces apoptosis in kidney cells of mice and activates PINK1/Parkin-mediated mitophagy. Parkin knockout mice showed more severe kidney damage caused by aluminum, suggesting that reduced mitophagy exacerbates oxidative stress and apoptosis in this context.

PINK1/TAX1BP1-directed mitophagy attenuates vascular endothelial injury induced by copper oxide nanoparticles

Yinzhen Fan et al.

Summary: This study found that CuONPs exposure disrupted mitochondrial dynamics in vascular endothelial cells through oxidative stress-dependent manner, as evidenced by increased mitochondrial fission and accumulation of fragmented mitochondria. Inhibition of mitochondrial fission aggravated CuONPs-induced mtROS production and cell death. Furthermore, mitochondrial fission led to the activation of PINK1-mediated mitophagy, and inhibition of mitophagy repressed CuONPs-induced toxicity. TAX1BP1 was identified as the primary receptor linking ubiquitinated mitochondria with autophagosomes, and its knockdown aggravated CuONPs-induced cell death. Urolithin A, a mitophagy activator, promoted mtROS clearance and removal of damaged mitochondria induced by CuONPs exposure both in vitro and in vivo.

Exercise triggers CAPN1-mediated AIF truncation, inducing myocyte cell death in arrhythmogenic cardiomyopathy

Stephen P. Chelko et al.

Summary: Research has shown that in a mouse model mimicking arrhythmogenic cardiomyopathy, myocardial cell death and heart dysfunction are primarily affected by calcium overload and oxidation of apoptosis-inducing factor (AIF). Preventing CAPN1-induced AIF truncation or inhibiting the binding of AIF to the nuclear chaperone PPIA may prevent myocardial cell death and slow down disease progression in ACM and other cardiomyopathies.

Upregulation of brain cholesterol levels inhibits mitophagy in Alzheimer disease

Vicente Roca-Agujetas et al.

Summary: Mitochondrial dysfunction in Alzheimer's disease is associated with impaired mitophagy due to accumulation of damaged mitochondria and high cholesterol levels. Cholesterol enrichment leads to disruption of mitophagy flux and cell aggregation of mitophagy adaptors, contributing to the progression of AD. Strategies focusing on restoring cholesterol homeostasis and mitochondrial redox balance may provide potential therapeutic options for AD.

Mitophagy protein PINK1 suppresses colon tumor growth by metabolic reprogramming via p53 activation and reducing acetyl-CoA production

Kunlun Yin et al.

Summary: Research has shown that the mitochondria protein PINK1 suppresses colon tumor growth by metabolic reprogramming and reducing acetyl-CoA production. PINK1 overexpression promotes mitophagy, decreases glycolysis, and increases mitochondrial respiration, while PINK1 deletion leads to decreased apoptosis, increased glycolysis, and reduced mitochondrial respiration.
