Characterization of polymer-polymer aqueous two-phase system droplets for 3D culture future applications

Summary: This study aimed to characterize the generation of polymer-polymer aqueous two-phase system (ATPS) droplets based on diameter size, morphology, and uniformity. Different ATPS compositions, construction strategies, and droplet phase volumes were evaluated. The results showed that the diameter size, morphology, and uniformity of ATPS droplets were influenced by ATPS compositions, construction strategies, and droplet phase volumes, with lower polymer bulk volumes resulting in higher uniformity. A uniform ATPS droplet with a mid-range diameter of 1.35 +/- 0.23 mm could be generated by immersing 1 mu L DEX droplets in a well filled with 50 mu L PEG.

Cellular Interaction of Bone Marrow Mesenchymal Stem Cells with Polymer and Hydrogel 3D Microscaffold Templates

Summary: In this study, three-dimensional scaffolds based on woodpile structures were fabricated using two-photon polymerization technology. The scaffolds were designed to mimic biological niches of healthy tissues or tumors. The effect of scaffold dimensions and mechanical properties on the adhesion and proliferation of bone marrow mesenchymal stem cells was investigated. The results suggest that the designed scaffolds and materials are suitable for further research in medical and pharmacological studies.

All-Aqueous Freeform Fabrication of Perfusable Self-Standing Soft Compartments

Summary: Compartmentalized structures obtained in all-aqueous settings have shown promising properties as cell encapsulation devices and reactors for trans-membrane chemical reactions. The interfacial stability conferred by emulsion systems enables the production of complex architectures. In this study, self-standing, malleable and perfusable tubular systems were fabricated through all-aqueous interfacial assembly, which can be used for cell culture and customized applications.

Summary: An aqueous two-phase system (ATPS) is a liquid-liquid phase separation system that shows great potential for various applications in the extraction and separation of biomolecules. Water-in-water emulsions have promising applications in the biomedical field. However, the low interfacial tension between the two phases makes it challenging to generate water-in-water emulsion droplets. Microfluidic platforms offer a solution by providing monodisperse emulsion droplets with a spherical shape, small volumes, and short reaction times.

A technique called aqueous two-phase systems in three-dimensional cultures has been proposed to address these challenges by promoting neural differentiation. Factors like cell density, colony size, interconnectivity, and substrate choice play important roles in determining neural differentiation efficiency in these cultures.
