Roles of the actin cytoskeleton in ciliogenesis

Huxley K. Hoffman et al.

Summary: Primary cilia are important for cells to respond to extracellular stimuli. Recent research has shed light on the regulation of ciliogenesis in different cell types. Actin cytoskeleton dynamics play a crucial role in various stages of ciliogenesis. Additionally, studies have revealed the requirements for actin remodeling in the formation of motile cilia in multiciliated epithelial cells, suggesting an alternative ciliogenesis pathway.

Microridge-like structures anchor motile cilia

Takayuki Yasunaga et al.

Summary: Motile cilia of Xenopus epidermis are anchored to microridge-like membrane protrusions to maintain their directionality. The actin cytoskeleton is dynamic during the early development of multiciliated cells, and subapical actin filaments are anchored to microridge-like structures. These observations identify microridge-like structures as an essential component of basal body rootlet anchoring in MCCs.

Summary: The article discusses the role of regulated airway epithelial development in the differentiation of motile ciliated cells in the human respiratory tract, as well as ciliary dysfunction caused by genetic mutations. Advances in understanding respiratory epithelial development and ciliary biology have the potential to improve disease diagnosis, management, and family counseling.

Control of multiciliogenesis by miR-34/449 in the male reproductive tract through enforcing cell cycle exit

Yu-Jie Wu et al.

Summary: The study showed that miR-34b/c and miR-449 play an important role in multiciliogenesis in embryonic development by regulating cell cycle exit. Depletion of miR-34b/c and miR-449 in a dKO mouse model led to a reduced number of multiciliated cells and abnormal cilia structure, which could be partially rescued by using a cyclin-dependent kinase inhibitor.

Fine-tuning multiciliated cell differentiation at the post-transcriptional level: contribution of miR-34/449 family members

Ioannis Loukas et al.

Summary: Cell differentiation towards a multiciliated cell fate is a tightly regulated process involving a transcriptional cascade and the formation of multiple centrioles and cilia. Micro-RNAs, particularly miR-34 and miR-449, play crucial roles in fine-tuning the expression of genes related to centrioles and cilia, ensuring proper commitment and maintenance of the multiciliated cell fate. Their spatiotemporal regulation during multiciliogenesis is essential for the coordinated movement of cilia and directional fluid flow across epithelia.
