Loss of physical contact in space alters the dopamine system in C. elegans

Surabhi Sudevan et al.

Summary: Progressive decline of neuromuscular function in microgravity is a persistent concern for interplanetary human habitation. This study reveals that impairments in the dopamine system are consistently observed in organisms exposed to spaceflight and simulated microgravity. Manipulation of the external environment through contact stimuli can prevent muscular changes and provide a realistic and viable treatment strategy for safe deep-space travel.

Role of oxidation of excitation-contraction coupling machinery in age-dependent loss of muscle function in Caenorhabditis elegans

Haikel Dridi et al.

Summary: The UNC-68 in C. elegans exhibits similar age-dependent dysfunction as mammalian RyR channels, and its oxidation and depletion of FKB-2 contribute to muscle Ca2+ leak and weakness.

Mitochonic Acid 5 Improves Duchenne Muscular Dystrophy and Parkinson's Disease Model of Caenorhabditis elegans

Xintong Wu et al.

Summary: Mitochonic Acid 5 (MA-5) enhances mitochondrial ATP production, restores fibroblasts from mitochondrial disease patients, and extends the lifespan of the disease model Mitomouse. MA-5 also interacts with mitofilin and modulates the mitochondrial inner membrane organizing system (MINOS) in mammalian cultured cells. In this study, researchers used Caenorhabditis elegans to investigate the effect of MA-5 on the Duchenne muscular dystrophy (DMD) model. The results showed that MA-5 improved the symptoms of the DMD model and protected against mitochondria perturbation. MA-5 also reduced the degeneration of dopaminergic cephalic (CEP) neurons and mitochondrial swelling in the Parkinson's disease (PD) model. These findings indicate that MA-5 may be a potential therapeutic agent for treating mitochondrial dysfunction in DMD and PD.

Mitochondrial hydrogen sulfide supplementation improves health in the C. elegans Duchenne muscular dystrophy model

Rebecca A. Ellwood et al.

Summary: Duchenne muscular dystrophy (DMD) is a genetic disorder characterized by muscle degeneration and weakness. Recent studies have shown that hydrogen sulfide (H2S) supplementation may improve muscle function and metabolism, suggesting a potential therapeutic approach for DMD. Deficits in H2S may contribute to DMD pathology, and delivery of H2S compounds could be a promising treatment strategy.

Pharmacological activation of SERCA ameliorates dystrophic phenotypes in dystrophin-deficient mdx mice

Ken'ichiro Nogami et al.

Summary: The study demonstrates that using SERCA activator CDN1163 can improve symptoms in dystrophin-deficient mice, prevent muscular damage, and enhance muscular strength. Continuous treatment for 7 weeks can reduce muscular degeneration and fibrosis. This research provides preliminary evidence for treating DMD by pharmacologically activating SERCA, and CDN1163 surprisingly improves muscular strength in wild-type mice.
