Summary: Gene therapy shows potential in curing cancer by editing tumor cells' deleterious genes, but inefficient delivery of genetic material into specific tumor sites limits its efficacy in cancer gene therapy. Lentiviral vectors (LVs) can be modified through pseudotyping to selectively kill tumor cells, as they have altered tropism and cell-type specificity in transduction. LVs' ability to efficiently deliver genes in vivo makes them attractive tools in future cancer gene therapy. This review summarizes the current status of pseudotyping LVs and their applications in basic anti-cancer studies across various malignancies, as well as discusses the opportunities of translating pseudotyped LVs into clinical use in cancer therapy.
