Does ventricle size contribute to cognitive outcomes in posthemorrhagic hydrocephalus? Role of early definitive intervention

Mounica Paturu et al.

Summary: In preterm infants with posthemorrhagic hydrocephalus (PHH), earlier permanent CSF (pCSF) diversion and smaller cumulative ventricle size were associated with larger right hippocampal volumes, improved neurocognitive outcomes, and reduced long-term ventriculomegaly. Future prospective randomized studies are needed to confirm these findings.

Summary: This study induced hydrocephalus in a pig model and found increased cell death in the periventricular white matter, decreased cell proliferation in the subventricular zone, and the occurrence of neuroinflammation.

Immune Regulation of Adult Neurogenic Niches in Health and Disease

Sana Chintamen et al.

Summary: Microglia play a crucial role in regulating neuronal development at different developmental stages and in specific environments of the adult brain. They modulate adult hippocampal neurogenesis by secreting factors that maintain a balance between cell proliferation and cell death, actively pruning synapses and engulfing excess neurons to shape the nervous system. Aberrant microglial activity in neurodegenerative diseases can impede the development of appropriate functional properties of adult born granule cells.

Summary: This study confirmed the efficacy of MSC-derived EVs in attenuating severe IVH-induced brain injuries, showing that the neuroprotection is primarily achieved through BDNF transfer via EVs, which is as effective as parental MSCs.

Cerebrospinal fluid biomarkers of neuroinflammation in children with hydrocephalus and shunt malfunction

Carolyn A. Harris et al.

Summary: The study found that non-obstructive cases of hydrocephalus had significantly higher levels of certain cytokines and MMPs in CSF compared to obstructive cases, suggesting a potential role of neuroinflammation in shunt failure. Factors such as patient age and number of past revisions were also found to be associated with shunt failure. Notably, elevated concentrations of IL-10, IL-6, IL-8, MMP-7, and MMP-9 were observed in non-obstructed cases.

TNF-mediated neuroinflammation is linked to neuronal necroptosis in Alzheimer's disease hippocampus

Anusha Jayaraman et al.

Summary: The underlying pathogenetic mechanisms of neuronal death and dysfunction in Alzheimer's disease are not fully understood, but chronic neuroinflammation is believed to play a role. The TNF superfamily of cytokines, known for their involvement in chronic inflammatory conditions, can activate neuroinflammatory responses that may lead to neuronal damage. This study shows that TNF-mediated necroptosis, rather than apoptosis, may be responsible for neuronal loss in AD, and that therapeutic intervention at various points in the signaling pathway may be possible.

Genetics and Molecular Pathogenesis of Human Hydrocephalus

Maria Garcia-Bonilla et al.

Summary: Hydrocephalus is a neurological disorder with a complex molecular pathogenesis that involves both genetic and environmental influences, affecting about 80-125 per 100,000 live births in the United States. From well-characterized heritable forms to complex pathologies like neural tube defects and Dandy-Walker syndrome, the genetic and molecular alterations in human hydrocephalus are varied. Key patterns among congenital and acquired forms of hydrocephalus include disruptions in cadherins, which play a role in neuroepithelium/ependymal cell junctions and contribute to the severity of the disease. Future research should focus on elucidating the genetic and molecular mechanisms regulating ventricular zone integrity and stem cell biology in relation to hydrocephalus pathogenesis and neurodevelopment.
