Epilepsy-associated increase in gonadotropin-releasing hormone neuron firing in diestrous female mice is independent of chronic seizure burden severity

Jiang Li et al.

Summary: This study demonstrates that the severity of epilepsy and the degree of disruption to GnRH neuron activity are independent, suggesting that susceptibility to reproductive endocrine disorders is driven by other risk factors.

Chronic androgen excess in female mice does not impact luteinizing hormone pulse frequency or putative GABAergic inputs to GnRH neurons

Chris S. Coyle et al.

Summary: Polycystic ovary syndrome (PCOS) is associated with androgen excess and abnormal luteinizing hormone (LH) secretion. Chronic exposure to androgens in mice leads to reproductive and metabolic impairments, but does not affect GABAergic input to GnRH neurons. These findings suggest that centrally driven mechanisms in hyperandrogen-mediated PCOS pathophysiology may vary depending on the developmental stage of androgen excess.

Phenotypic differences based on lateralization of intrahippocampal kainic acid injection in female mice

Cathryn A. Cutia et al.

Summary: Clinical evidence suggests that there are differential outcomes of comorbid conditions in patients with temporal lobe epilepsy (TLE) based on the lateralization of the seizure focus. This study evaluated the effects of targeting left or right dorsal hippocampus in a model of TLE, and found that it produced different outcomes in granule cell dispersion, body weight gain, and reproductive endocrine dysfunction in female mice. The findings suggest that lateralization of the epileptogenic insult may contribute to similar reproductive endocrine outcomes.

Increased GABA transmission to GnRH neurons after intrahippocampal kainic acid injection in mice is sex-specific and associated with estrous cycle disruption

Robbie J. Ingram et al.

Summary: Patients with epilepsy have a higher rate of reproductive endocrine comorbidities. This study found that GnRH neuron firing is increased in IHKA mice with disrupted estrous cycles, and this is associated with an increase in GABAergic transmission. Additionally, there is a sex-specific hypothalamic mechanism underlying reproductive endocrine dysfunction in IHKA mice.

Development of a Highly Sensitive ELISA for Measurement of FSH in Serum, Plasma, and Whole Blood in Mice

Luisina Ongaro et al.

Summary: The article introduces a novel and sensitive ELISA that enables accurate measurement of FSH in serum, plasma, and whole blood from female and male mice. The assay can also be used to measure FSH in murine pituitary lysates and cell culture media. This new methodology allows investigators to measure FSH in mice from various biological samples accurately, at low cost, and in their own laboratories.

Modulation of pulsatile GnRH dynamics across the ovarian cycle via changes in the network excitability and basal activity of the arcuate kisspeptin network

Margaritis Voliotis et al.

Summary: Research suggests that KNDy neurons play a crucial role in regulating pulsatile release of GnRH through modulation of neuropeptide and glutamate signaling. Optogenetic stimulation of KNDy neurons stimulates pulsatile release in estrous mice but inhibits it in diestrous mice. This highlights the importance of network excitability in pulse generation and may have implications for reproductive therapy targeting the glutamate system.

Impact of chronic variable stress on neuroendocrine hypothalamus and pituitary in male and female C57BL/6J mice

Betina B. Nair et al.

Summary: Chronic stress can have multiple negative effects on an individual's physiology and health, particularly affecting female reproductive function. However, most neuroendocrine axes seem to be resilient to the effects of chronic stress.

The dendron and episodic neuropeptide release

Allan E. Herbison

Summary: The dendron is a unique structure of GnRH neurons that integrates afferent information and generates pulsatile output. Proximal dendrites receive synaptic inputs, while distal dendrons regulate GnRH secretion through electrotonic propagation.
