Effective drug combinations in breast, colon and pancreatic cancer cells

Patricia Jaaks et al.

Summary: Combinations of anti-cancer drugs can overcome resistance and provide new treatments. This study evaluates 2,025 clinically relevant two-drug combinations and finds that synergy between drugs is rare and context-dependent. The findings can guide the development of combination treatments for specific cancer subtypes.

Mechanisms of antibiotic action shape the fitness landscapes of resistance mutations

Colin Hemez et al.

Summary: Antibiotic-resistant pathogens pose a significant public health threat. Understanding how the mechanism of action of antibiotics influences the emergence of resistance is crucial for the development of new drugs and the preservation of the effectiveness of existing ones. The study found that both bacteriostatic and bactericidal agents can effectively suppress the selection of resistant mutants. Additionally, heterogeneous drug-target binding within a population enables resistant bacteria to evolve fitness-improving secondary mutations.

The distribution of cellular turnover in the human body

Ron Sender et al.

Summary: The research integrated ubiquity, mass, and lifespan of major cell types to quantitatively describe cellular turnover, finding a total cellular mass turnover of 80+/-20 grams per day, primarily from blood cells and gut epithelial cells. The study also revealed that close to 90% of the daily cell turnover was from blood cells, providing an estimation of the distribution of cellular turnover in the human body.

RESTAMP - Rate estimates by sequence-tag analysis of microbial populations

Anel Mahmutovic et al.

Summary: Microbial division rates and death rates play a crucial role in the evolution of antimicrobial resistance, and measuring these rates can enhance our understanding of host-pathogen interactions and antibiotic action. RESTAMP is a method that utilizes fitness neutral sequence barcodes to measure bacterial division and death rates, correcting for technical bottlenecks in the process.

The stagnation paradox: the ever-improving but (more or less) stationary population fitness

Hanna Kokko

Summary: Fisher's fundamental theorem suggests that natural selection improves mean fitness, but the measurement of population fitness is not clear, leading to the stagnation paradox. With a deeper understanding of these concepts, the paradox disappears, and adaptation can result in a decline in population reproductive output, especially when individuals can improve their fitness by exploiting conspecifics.

The impact of phenotypic heterogeneity of tumour cells on treatment and relapse dynamics

Michael Raatz et al.

Summary: Within a cancer cell population, individual cells are not identical, leading to heterogeneity that affects disease progression and treatment success. Adjusting treatment based on growth rate can delay relapse but may reduce overall treatment effectiveness. Combining different treatment types can increase efficiency and delay relapse.

Distinct clonal lineages and within-host diversification shape invasive Staphylococcus epidermidis populations

Anna Both et al.

Summary: S. epidermidis demonstrates multifactorial processes of infection adaptation, including intra-clonal heterogeneity, mutations and deletions in specific genes, and enhanced biofilm formation. This adaptive process allows invasive S. epidermidis to survive in hostile environments and progress to chronic implant-associated infections.

High potency of sequential therapy with only β-lactam antibiotics

Aditi Batra et al.

Summary: Research found that rapid switching between β-lactam antibiotics increased bacterial population extinction; low rates of spontaneous resistance emergence and low levels of spontaneous cross-resistance supported this extinction phenomenon. These findings help guide the optimized use of available antibiotics in treatment designs.
