Prognostic and therapeutic significance of a novel ferroptosis related signature in colorectal cancer patients

Songtao Du et al.

Summary: This study constructed a prognostic model for colorectal cancer (CRC) by analyzing gene data and clinical information, and provided an alternative individualized treatment strategy. Specific genes related to the prognosis of CRC were identified, and enrichment analysis determined the signaling pathways associated with these genes, providing new targets for treatment. In addition, a drug was screened and its combination with specific genes was found to significantly enhance the treatment effect.

Down-regulating Nrf2 by tangeretin reverses multiple drug resistance to both chemotherapy and EGFR tyrosine kinase inhibitors in lung cancer

Ying Xie et al.

Summary: This study investigates the potential of targeting the Nrf2 pathway to overcome acquired drug resistance and enhance the therapeutic effects of chemotherapy and molecular-targeted therapy. It demonstrates that upregulation of Nrf2 in Gefitinib-resistant and paclitaxel-resistant lung cancer cells is an important mechanism.

SIRT6 silencing overcomes resistance to sorafenib by promoting ferroptosis in gastric cancer

Shunv Cai et al.

Summary: This study found that silencing SIRT6 can overcome sorafenib resistance in gastric cancer by promoting ferroptosis. Mechanistically, SIRT6 inhibition leads to inactivation of the Keap1/Nrf2 pathway and downregulation of GPX4, while over-expression of GPX4 or activation of Keap1/Nrf2 can reverse the effects. Targeting the SIRT6/Keap1/Nrf2/GPX 4 pathway may be a potential strategy for overcoming sorafenib resistance in gastric cancer.
