Very-Long-Chain Unsaturated Sphingolipids Mediate Oleate-Induced Rat β-Cell Proliferation

Anne-Laure Castell et al.

Summary: This study found that oleate can induce the proliferation of pancreatic beta cells through the production of very-long-chain unsaturated sphingolipids in the presence of high glucose. Inhibiting key enzymes or proteins in the sphingolipid synthesis pathway can reduce the effect of oleate on beta cell proliferation.

CB1 Cannabinoid Receptor Signaling and Biased Signaling

Luciana M. Leo et al.

Summary: The CB1 cannabinoid receptor is a promising therapeutic target for various disorders, but the development of drug candidates is hindered by challenges such as adverse effects and rapid tolerance. Biased signaling analysis may help dissociate therapeutic and adverse effects, but the mechanism of biased signaling at the CB1 receptor remains poorly understood at present.

Functional Selectivity of a Biased Cannabinoid-1 Receptor (CB1R) Antagonist

Ziyi Liu et al.

Summary: The study found that the peripheral CB1R antagonist MRI-1891 is highly biased towards inhibiting CB1R-induced beta-arrestin-2 activity, reducing food intake and body weight without eliciting anxiety while improving muscle insulin resistance. In contrast, the global CB1R antagonist rimonabant may cause anxiety and does not involve beta-arrestin-2.

Peripherally Selective CB1 Receptor Antagonist Improves Symptoms of Metabolic Syndrome in Mice

Nayaab Khan et al.

Summary: The study demonstrates that the CB1 antagonist RTI1092769, with limited brain exposure, can improve MetS-related complications such as weight gain, glucose utilization, and hepatic triglyceride content. This suggests that CB1 antagonists with restricted CNS effects may be beneficial for treating MetS and related conditions.
