Encapsulation of doxorubicin prodrug in heat-triggered liposomes overcomes off-target activation for advanced prostate cancer therapy

Summary: In this study, the encapsulation of Dox-PSA into LTSL was successfully achieved, allowing controlled release upon temperature increase and improving the safety and biological activity of Dox-PSA. Efficient release, cleavage, and nuclear entry of Dox-PSA were observed in prostate cancer cells. Furthermore, Dox-PSA-loaded LTSL showed prolonged blood circulation, reduced cardiac toxicity, and improved therapeutic efficacy compared to free Dox-PSA. Lastly, the combination of Dox-PSA-loaded LTSL and HT delayed tumor growth, providing a safer option for the treatment of locally advanced prostate cancer.

Synthesis of a targeted, dual pH and redox-responsive nanoscale coordination polymer theranostic against metastatic breast cancer in vitro and in vivo

The developed intelligent platform demonstrated high drug loading capacity, pH and redox-responsive drug release, and showed outstanding therapeutic and diagnostic capabilities against breast cancer.

Targeted biomimetic hollow mesoporous organosilica nanoparticles for delivery of doxorubicin to colon adenocarcinoma: In vitro and in vivo evaluation

Summary: In this study, hollow mesoporous organosilica nanoparticles capped with red blood cell membrane were designed to improve the therapeutic efficacy of drugs through GSH-responsive doxorubicin releasing mechanism. A targeted system was prepared using a DNA aptamer against MUC-1 receptor for guided delivery of doxorubicin to colorectal tumor cells. Experimental data showed that the aptamer facilitated drug uptake and the targeted system exhibited enhanced tumor targeting capability in a mouse model. The GSH-responsive system also reduced drug toxicity and improved survival rate, suggesting its potential in cancer therapy.

Synthesis of doxorubicin-loaded peptosomes hybridized with gold nanorod for targeted drug delivery and CT imaging of metastatic breast cancer

Summary: In this study, polypeptide-based vesicles called peptosomes were prepared using synthetic amphiphilic polypeptide-based PEG-PBLG diblock copolymer, and were successfully used for co-delivery of doxorubicin and gold nanorods. The results showed that the targeted peptosomes exhibited higher cellular uptake and cytotoxicity, and demonstrated good performance in both therapy and diagnosis.
