The effect of buffer species on biorelevant dissolution and precipitation assays-Comparison of phosphate and bicarbonate buffer

Summary: Biorelevant solubility and dissolution testing is an important tool in pharmaceutical development. However, the use of biorelevant media does not always match the solubility data observed in human intestinal fluids. This study found that the medium primarily affects dissolution kinetics, and the unique buffering properties of bicarbonate buffer play a special role in simulating conditions in human intestinal fluids. Further investigation is needed to understand the factors causing differences in solubility and dissolution behavior between phosphate- and bicarbonate-buffered biorelevant media.

Cocrystals of Nitrofurantoin: How Coformers Can Modify Its Solubility and Permeability Across Intestinal Cell Monolayers

Summary: This study compared the solubility and permeability of nitrofurantoin with its cocrystals and mixtures containing different coformers. It was found that the solubility of nitrofurantoin was reduced when mixed with phenanthroline and bipyridyl, but not with isoniazid. Molecular dynamics simulations were used to explain these experimental data based on nitrofurantoin-coformer and nitrofurantoin-water interactions. Permeation studies showed that the nitrofurantoin-phenanthroline mixture was detrimental to the monolayer integrity, while the cocrystal had no effect. Nitrofurantoin permeability and monolayer integrity were not affected by the nitrofurantoin-isoniazid or nitrofurantoin-bipyridyl mixtures.

Apigenin-oxymatrine binary co-amorphous mixture: Enhanced solubility, bioavailability, and anti-inflammatory effect

Summary: This study aimed to improve the bioavailability of APG by a food-friendly co-amorphous formulation of APG with oxymatrine (OMT). Results showed that the complexation between APG and OMT significantly enhanced the solubility and dissolution of APG-OMT CM, leading to higher oral bioavailability and anti-inflammatory effects compared to pure APG. The study provides a promising delivery system for improving the bioavailability of hydrophobic food ingredients.
