Electrochemical Synthesis of Pyrazolines and Pyrazoles via [3+2] Dipolar Cycloaddition

Martin Linden et al.

Summary: This study presents the first electrochemical approach for the direct synthesis of pyrazolines and pyrazoles from easily accessible hydrazones and dipolarophiles. The method allows for large-scale production and even conversion of highly sensitive alkenes. The use of a biphasic system with sodium iodide as a supporting electrolyte and mediator is cost-effective. Mechanistic insight is provided by the isolation of key intermediates, and the synthesis of a commercial herbicide safener further highlights the significance of this reaction.

Phenols in Pharmaceuticals: Analysis of a Recurring Motif

Kevin A. Scott et al.

Summary: Phenols and phenolic ethers are important molecular structures in nature and approved drugs. This study provides the first comprehensive compilation and analysis of the structures of U.S. FDA-approved molecules containing phenol or phenolic ether fragments. The research reveals the differences in physicochemical properties between phenols and phenol ethers, and their significance in drug development. Despite recent trends in increasing sp(3) centers in drug leads, phenols and phenolic ethers still have a significant presence in small-molecule drugs approved in 2020, indicating their special role in drugs and natural products.

Robust and Self-Cleaning Electrochemical Production of Periodate

Camila M. Kisukuri et al.

Summary: Periodate, a platform oxidizer, can be recycled through electrochemical re-oxidation in the presence of organic impurities. Cold combustion of organic compounds to CO2 and H2O provides orthogonal conditions for periodate synthesis. This strategy can be used to degrade organic contaminants and control the purity of periodate.

Electrosynthesis of 2-Substituted Benzoxazoles via Intramolecular Shono-Type Oxidative Coupling of Glycine Derivatives

Lei Liu et al.

Summary: Herein, an atom-economical and eco-friendly electro-chemical oxidation/cyclization of glycine derivatives through intramolecular Shono-type oxidative coupling is disclosed, leading to a variety of 2-substituted benzoxazoles in yields of 51-85%. This oxidative cyclization proceeded in transition metal-and oxidant-free conditions and generated H2 as only a byproduct. Additionally, gram-scale reactions and a broad substrate scope demonstrated the synthetic usefulness of this protocol.

Reactor Design for the Direct Electrosynthesis of Periodate

Sebastian Arndt et al.

Summary: This article presents a reactor design for continuously producing sodium paraperiodate under strongly alkaline conditions. The design combines the concepts of cyclic flow electrolysis and a continuous stirred tank reactor (e-CSTR), allowing for fully continuous operation and the use of various iodine sources. Further developments include the use of dissolved or solid starting materials, inline separation of the product, and recycling of electrolytes, enabling cost-efficient synthesis of metaperiodate in Ph.Eur. quality.

Electrochemical synthesis of N,N′-disubstituted indazolin-3-ones via an intramolecular anodic dehydrogenative N-N coupling reaction

Jessica C. Bieniek et al.

Summary: The use of electricity as a traceless oxidant provides a sustainable and novel approach to synthesize N,N'-disubstituted indazolin-3-ones through an intramolecular anodic dehydrogenative N-N coupling reaction. This method offers mild reaction conditions, easy experimental setup, excellent scalability, and high atom economy. Various indazolin-3-one derivatives were synthesized with yields up to 78% using inexpensive and sustainable electrode materials and a low supporting electrolyte concentration. Mechanistic studies suggest a biradical pathway and the cleavage of protecting group can be used to access single 2-aryl substituted indazolin-3-ones.
