Recurrent mutations in topoisomerase IIα cause a previously undescribed mutator phenotype in human cancers

Arnoud Boot et al.

Summary: This study identified a mutation in human topoisomerase hTOP2 alpha (p.K743N) and linked it to a previously undescribed mutator phenotype in cancer. The research determined the genetic requirements and mechanisms of this mutagenic process and showed that it may contribute to tumorigenesis. The results shed further light on the role of topoisomerase II in genome instability.

Topoisomerase II Poisons: Converting Essential Enzymes into Molecular Scissors

Kendra R. Vann et al.

Summary: The complex and restricted movement of DNA in eukaryotic cells leads to various topological issues that impact all genetic functions. Topoisomerases regulate DNA topology and play crucial roles in processes such as DNA replication, transcription, and recombination. However, their ability to generate double-stranded breaks in the genetic material makes type II topoisomerases inherently dangerous enzymes, which are targeted by many anticancer drugs.

Structural basis for allosteric regulation of Human Topoisomerase IIα

Arnaud Vanden Broeck et al.

Summary: Human type IIA topoisomerases play a crucial role in regulating DNA topology and chromosome organization. The isoform Topo II alpha is a key target for anti-cancer compounds, forming ternary cleavage complexes with DNA. This study provides cryo-EM structures of the entire Topo II alpha nucleoprotein complex, shedding light on the allosteric connections between different domains and the role of the non-conserved C-terminal domain in regulating enzyme activities.
