Towards understanding single-channel characteristics of OccK8 purified from Pseudomonas aeruginosa

Fatma Dogan Guzel et al.

Summary: This study investigates a substrate-specific channel OccK8 from Gram-negative bacteria, showing multiple conformations in lipid bilayers. The research suggests that positively charged arginine interacts infrequently with OccK8, while negatively charged glutamic acid preferentially interacts with the channel.

Rapid fabrication of teflon apertures by controlled high voltage pulses for formation of free standing planar lipid bilayer membrane

Yasin Ozturk et al.

Summary: The study investigates the reproducible generation of micropores in the range of 50-100 microns in diameter in a Teflon film. The multi-pulse method is more convenient for lipid bilayer formation and has a higher success rate. Experimental results show that the lipid bilayer on Teflon pores can last for more than 33 hours.

On-chip label-free impedance-based detection of antibiotic permeation

Jaspreet Kaur et al.

Summary: This article introduces a microelectrode integrated microfluidic device for rapid and label-free detection of antibiotic permeation across a membrane, enabling the recognition of antibiotic permeation through electrical impedance measurement and optical monitoring. Results show that the proposed integrated device can easily monitor the permeation of antibiotic molecules, providing a clear distinction between bilayer and porin-mediated permeation through lipidic bilayer and porin channels.

How to Enter a Bacterium: Bacterial Porins and the Permeation of Antibiotics

Jigneshkumar Dahyabhai Prajapati et al.

Summary: Traditional antibiotics are facing the challenge of Gram-negative bacteria developing resistance, prompting the urgent need for new antibacterial agents. Designing new antibiotic molecules that can rapidly penetrate Gram-negative bacteria and achieve lethal intracellular drug accumulation is a major challenge. The lack of simple experimental techniques to measure antibiotic uptake and local concentrations in subcellular compartments is a limiting factor.

Towards the sustainable discovery and development of new antibiotics

Marcus Miethke et al.

Summary: There is a growing demand for novel antimicrobials to treat infections caused by multidrug-resistant bacterial pathogens, but there is currently a lack of investment in this field. New agents with innovative chemistry and modes of action are urgently needed to combat the public health threat posed by antimicrobial resistance. The consortium proposes short-term and long-term solutions to overcome urgent limitations in research and funding, aiming to bridge the gap between academic, industrial, and political stakeholders to improve the ability to discover and develop new antibiotics.
