Cisplatin nephrotoxicity: new insights and therapeutic implications

Summary: The authors review the mechanisms of cisplatin-induced kidney injury and discuss the challenges and potential targets for renoprotection. The cellular and molecular mechanisms of cisplatin nephrotoxicity involve DNA damage, mitochondrial pathology, oxidative stress, endoplasmic reticulum stress, and stress responses such as autophagy, cell-cycle arrest, senescence, apoptosis, programmed necrosis, and inflammation. Emerging evidence suggests a contribution of epigenetic changes to cisplatin-induced kidney injury. Potential therapeutic targets have been identified, but their effects on the efficacy of cisplatin chemotherapy need further evaluation.
